Heavy metal is widely spreaded both in the earth crust and water environment. Undernatural conditions, trace amounts of heavy metals generally do not affect human health.However, the development of the human society increased the production of heavy metals.Nowadays, heavy metals are widely used in industry, medicine, etc. In improving human lifeat the same time also caused a large number of heavy metals into the environment andinduced to the environment pollution. The heavy metal pollution can be accumulated throughfood chain. It is hard to be removed but transfer. Monitoring of heavy metals in theenvironment has caused widespread attention.The usual detecting methods for heavy metal are AES, AAS, AFS and MS. Thetraditional detection methods for operating personnel professional demand is higher, and theequipments are not portable. In recent years, the biosensor technology based on nucleic acidprobe of heavy metals, due to the advantages of rapid response and convenient operation, hasgood application prospect.This paper introduces fluorescence detection methods for Ag+and Hg2+via the DNAaptamer. Where there is Ag+/Hg2+and the single strand DNA(ssDNA) aptamer there will bethe mismatch combination of C-Ag+-C/T-Hg2+-T producing the double strand DNA(dsDNA)structure. we have know that the fluorescence dye can insert the dsDNA structure and makeout strong fluorescence. The fluorescence intensity grows stronger when the Ag+/Hg2+reacting with the aptamer forming the dsDNA structure. It is easy to detect Ag+and Hg2+through fluorospectro photometer.In order to aquire accurate test results, we optimize the detection procedures. In purposeof examining the applicability, we studied the influence of common interferences in aqueoussolution to this detecting system. Studies have shown that there is little impact on this system.And the result of actual water samples detection is acceptable.
