Objective (1) To study the changes of scutellarin of Seutellariae Barbata Herba under different immersion conditions; (2) To study the extraction process of scutellarin from Seutellariae Barbata Herba for large-scale production; (3) To study the preparation of compound Banzhilian tablet for optimizing the best process condition and establishing the intact tablet formulation and the preparation process.Methods (1) HPLC was adopted to detect the content of scutellarin in the water extraction of Seutellariae Barbata Herba immersed in different temperature and time; the content of scutellarin (C) and time (t) were fitted by dynamic equation and the kinetics parameters were calculated; (2) Two parameters including alcohol concentration and addition method of the raw herb were investigated; (3) Crushing and sterilization processes was studied with the percentage of comminution and the content of berberine as an index,while the orthogonal experiment was used to select the optimal water extraction process with the content of scutellarin and total solid as an index;To investigate concentration and drying process with the content of scutellarin as an index and choose the best preparation process with hygroscopicity, angle of repose, appearance of tablets, disintegration time and hardness as an index.Results (1) Under the conditions of immersing between 20℃and 60℃, the changes of scutellarin significantly degraded in the water extraction of Seutellariae Barbata Herba, and degradation rate was fastest between 30℃and 40℃; While the content of scutellarin of water extraction of Seutellariae Barbata Herba kept invariant and it had a higher extraction rate under the conditions of immersion temperature above 70℃; (2) Both two parameters significantly affected the extracting efficiency. Seutellariae Barbata Herba extracted by boiling the raw herb in 50% alcohol was the optimal extraction, and the extracting efficiency of boiling in water was close to the optimal one; (3) The experimental results showed that the best preparation of compound Banzhilian tablet was as follows:Coptidis Rhizome and Notoginseng Radix et Rhizoma was crushed into fine powder;including Seutellariae Barbata Herba,the other eight herbs were extracted by 6-times volμme water for twice and a half-hour for each,it should add after boiling water for Seutellariae Barbata Herba,then, the filtrate was concentrated to plaster(the relative density was 1.25,25℃) after filtered and combined, it was mixed with powder and vacuμm dried and added appropriate amount of starch and dextrin after crushed into fine Powder,last, after making into particles and adding the right amount of magnesiμm stearate, it was compressed into 1000 tablet.Conclusion (1) Scutellarin of Seutellariae Barbata Herba existed enzymatic degradation reaction in the process of immersion; it can effectively increase the extraction of scutellarin rate while feeding in water with the temperature above 70℃; (2) The experiment indicates that Seutellariae Barbata Herba contains endogenous scutellarin enzymes.The method of boiling the raw herb in water can be applied into large-scale production of scutellarin from Seutellariae Barbata Herba, and has the advantage of lower input, simple operation and high safety coefficient; (3) As a sμmmary,the manufacture technology establish in the experiment was stable and practical,it ensured the scaled produation of the industrial production of Compound Banzhilian Tablet.
