Cloning And Function Analysis Of The Anthocyanin Biosynthesis Related Genes DFR And MYB In Eggplant Sepals

Eggplant (Solanum melongena L.) as the mass table is the common dish, which is rich in a lot of anthocyanins, have a high nutritional value, its roots, stems, leaves and seeds have some medicinal value, its sepals are often appeared on the table, it has good market prospects.This study is aimed at the observation of Europe Lve eggplant, Northeast purple eggplant, and they hybrid eggplant, Europe imports eggplant sepals are green throughout the growing cycle, the northeast purple eggplant sepals gradually goes all purple in its growth cycle.The hybrids with the growth cycle progresses, sepals gradually turned purple, but not all of purple, it’s purple-and-green-and-white. In this study, we made a research and results analysis for the reason for the changes of the sepals:1. Chose the grown ripe fruit sepals as experimental material. the green speals eggplant of Europe, the purple speals eggplant of northeast, and their hybrids which was green-and-purple-and-white. Clone the three kinds of eggplant’s DFR and MYB genes by using RACE and homologous cDNA clone, and named them as SmDFR and SmMYB.2.DFR and MYB analysisThe Brigitte RZ F1 Hybrids’ SmDFR cDNA length of 1016bp, from 82bp-716bp are open reading frame, encoding 213 nucleotides, named as KX224250;the LONGQIEYIHAO SmDFR cDNA length of 1128bp, from 332bp-973bp are open reading frame, encoding 213 nucleotides,named as KX224251;the Hybridization of Brigitte RZ F1 Hybrids and LONGQIEYIHAO SmDFR cDNA length of 1005bp, from 73bp-708bp open are reading frame, encoding 211 nucleotides, named as KX224252. The Brigitte RZ F1 Hybrids’ SmMYB cDNA length of 969bp, from 116bp-578bp are open reading frame, encoding 153 nucleotides,named as KX224253; the LONGQIEYIHAO SmMYB cDNA length of 959bp, from 178bp-621bp are open reading frame, encodingl53 nucleotides,named as KX224254; the Hybridization of Brigitte RZ F1 Hybrids and LONGQIEYIHAO SmMYB cDNA length of 993bp, from 140bp-582bp are open reading frame, encoding of 153 nucleotides,named as KX224255.3. Acquisition sepals, three varieties of anthocyanin content was measured four times. Test results show that the European greed speal varieties anthocyanin showed no significant change; and the Northeast Grimmia advance with the growth cycle, the anthocyanin content increased significantly; anthocyanin content of the hybrids in between the two, and purple sepals observed a positive correlation degree of change. Fluorescence quantitative PCR results showed that the expression levels of anthocyanin content of DFR and its MYB positive correlation.It proved that three varieties of colors and sepals anthocyanin content variation of DFR and MYB expression patterns and expression levels positively correlated.4.
