To reveal the mechanism of genetic variations induced by space radiation on plant seeds, dry seeds of rice after being carried by Shenzhou 6 to the space in 2005 were planted on the ground. Purple leaves mutants were observed in YG, L1 and L2, three different varieties of rice; and green leaves plants were observed in the next different generations of purple leaves mutants. It indicates that changes of rice leaf color are induced by space radiation via inducing genomic instability. For further investigating the mechanism of genomic instability induced by space radiation, we analyzed the transposon mPing sites polymorphism of the fifth generation of YG, L1 and L2 purple mutants via transposon differential display technique. These results indicate that the transposon mPing activation degrees of purple mutants are all significantly higher than green plants (p<0.05), except L1E5.It showed that the transposon mPing of the purple and green plants is active and the mPing activity of the purple plant is higher than that of the green plant.Meanwhile, the transposon mPing sites activation discrepancy rates of the fifth generation green and purple mutants are extremely higher (p<0.01). But the activation degrees of L1E5 is different. It is relatively stable. The results reveal that purple rice leaf caused by Space radiation is associated with activation degrees of transposon mPing.
