Nitrogen pollution is one of the main factors of the eutrophication, ammonia is an important component which cause nitrogen pollution of the water environment. The harm caused by eutrophication is varied. At present, all countries attach great importance to the management of eutrophication. China has chosen ammonia as a major indicators of water pollution in the "12th Five-Year" period. Microbial denitrification is the main biological methods of controling the eutrophication of water. Therefore, the screen of high ammonia nitrogen bacteria also arise people’s attention gradually.Bacteria which can remove ammonia were selected from samples collected in wetland plant rhizosphere soil. We take two separation schemes in the experiments. One method:we screened bacteria in general ammonia concentration medium, and gained 101 strains of bacteria. Another method:we screened bacteria in three different high ammonia concentration mediums and obtained 101 strains.The concentration of ammonia nitrogen mediums were 500mg/L,800mg/L and 1000 mg/L, and we isolated 86 strains,40 strains and 25 strains respectively.Filter isolated bacteria, the bacterial deamination efficiency filter the ammonia concentration in the medium, respectively, for the 500 mg/ L,800mg/L and 1000 mg/L. The effect of ammonia nitrogen removal of bacteria can be detected by Nessler’s reagent method, we gained two strains of ammonia removal bacteria L26 and Lh5 through the initial screen and second screen, and the effect of L26 and Lh5 ammonia removal were 20% and 18%.Compaired to a application of commercial bacteria, the ability of removal ammonia nitrogen from wastewater of L26 bacteria is better, while LH5 bacteria is worse. According to the above cases, we select L26 bacteria for further research. Morphological observation and 16SrDNA sequencing of L26 were implementated, we can concluded that L26 belong to Pseudomona by contrast.We analysed the influence removal ammonia of L26 bacteria by the growth temperature, inoculum size, pH and biomass.The results showed that the ability of removal ammonia of L26 bacteria was higher when L26 bacteria were grown at 30° C, grown in a condition of 1% inoculum for 4h, grown in pH 7 or 8, grown in log phase.
