Biogenic amines have a major impact on plant and animal physiological functions, But ifexcessive biogenic amines is intaked,it is poisoning, pathogenic even death.Therefore thecontent of biogenic amines has become an important indicator of food safety testing.Biogenicamine oxidases oxide biogenic amines to aldehydes as the main metabolite,and aldehydes willbe further oxidized to a carboxylic acid or the use of in vivo and eventually out of the body.Biogenic amine oxidases to reduce the content of biogenic amines and that is meaningful toensuring food safety. This study is about the biogenic amine oxidase produced byLactobacillus rhamnosus. The main contents are as follows:1.Researching the optimal training conditions of an biogenic amine oxidases produced byLactobacillus rhamnosusT TUsing single factor and orthogonal experimental study of Lactobacillus rhamnosusculture conditions to produce the most biogenic amines oxidases, cruding biogenic aminesoxidases and measuring its activity by spectrophotometry. The experiment got the besttraining conditions: incubation time 16 h, culture temperature 39 ℃, the carbon source isglucose, nitrogen source for yeast, initial p H of 7.5.2.Optimizing extract conditions of biogenic amine oxidase by PEG/(NHB4B)B2BSOB4Baqueous-phase systemsPEG/(NHB4B)B2BSOB4Baqueous-phase systems was used to extract biogenic amine oxidase thatproduced by Lactobacillus rhamnosus. Single factor experiment with PEG molecular massand concentration, ammonium sulfate concentration, p H value and temperature, Preliminarydetermine the optimum, then designed and analysed it with Box-Behnken experiment. Theresult indicated that PEG molecular mass was 2000, PEG2000 concentration was25%,ammonium sulfate concentration was 10%, p H value was 7.5 and the temperature was30 made of the best experiment conditions. Under above conditions,the extraction rate was96.5% and the purification factor was 2.38.3.Comparing the biogical oxidation of biogenic amine oxidase before and after extraction andobserving the he effect of additives on its oxidationT TComparing the oxidation of biogenic amine oxidase to the eight common biogenicamines before and after extraction by HPLC.After extraction, biogenic amine oxidaseenriched, enhanced oxidative capacity, but because of purification factor is not high, it did notachieve the desired effect oxidation. Adding glucose oxidation almost no effect on thebiogenic amine oxidase;adding sodium chloride inhibit the oxidation of biogenic amineoxidase,Which has strong inhibition to the putrescine, tryptamine, phenylethylamine,cadaverine, tyramine and spermidine, almost no effect on histamine and spermine.
