[Objective] In order to investigate the relationship between JAK-STAT signal pathway and Tibial dyschondroplasia (TD) in broiler chicken, this experiment was conducted to study on sequential expression of p-STAT3、SOCS3 genes in TD development in chickens.[Methods] This experiment employed the Real-time qPCR, immunohistochemical technique to observe the expression changes of p-STAT3 and SOCS3 genes in transcriptional and translation levels by constructing TD models in broiler chicken at the early stage.[Results]Compared to control group, the expression of STAT3 in the growth plate shows no difference (P>0.05) in thiram-fed group on day 1, however on days 2,4 and 6, the expressions presented an extremely significant differences (P<0.01); and the levels of SOCS3 gene expression in thiram-fed group was increased on days 1,2 and 6 with highly significant differences (P<0.01). It showed lower expression levels on day 4, the difference was significant (P<0.01).P-STAT3 protein expression located at the proliferative zone, prehypertrophic zone and hypertrophic zone of the growth plate. Compared with the control group, P-STAT3 protein on proliferative zone in the thiram-fed group was observed in over-expression on days 1,2 and 4, and down-regulated expression on day 6, which were highly significant difference (P<0.01) on days 1 and 2, and was significantly different on days 4 and 6 (P<0.05). p-STAT3 on the prehypertrophic zone and hypertrophic zone were up-regulated on days 1,2 and 4, and down-regulated on day 6 with highly significant differences (P<0.01); SOCS3 protein on proliferative zone of thiram-fed group was observed over-expressed on days 1,4 and 6, however its expression was found down-regulated on day 2, with no significant difference on days 1 and 6 (P>0.05), and were highly significant different on days 2 and 4 (P<0.01). SOCS3 protein on prehypertrophic zone was over-expressed on days 1,4 and 6, and down-regulated of expression on day 2, which with highly significant differences on days 1 and 4 (P<0.01), and was significant on day 2 (P<0.05), and no significant difference on day 6 (P>0.05). SOCS3 protein on the hypertrophic zone was over-expressed on days 1,2,4 and 6, and down-regulated expression on day 2, which with highly significant differences on days 1, 2 and 4 (P<0.01), and no significant difference on 6 (P>0.05).[Conclusions] The up-and down-regulation of STAT3 and SOCS3 were in parallel with gene microarray in the transcriptional level. P-STAT3 and SOCS3 expression located at the proliferative zone, prehypertrophic zone and hypertrophic zone of the growth plate. The differential expression of p-STAT3 and SOCS3 showed that JAK-STAT signaling pathway might play an important role in regulating an abnormal proliferation, differentiation or apoptosis of chondrocytes in TD at early stage.
