With the development of science and technology, research and utilization of marine microbes more and more people’s attention, marine microorganisms have unique characteristics, which make them easier to use by people. With the increase of people’s demand for collagen study collagenase gradually depth, microbial production of conventional extraction collagenase is relatively small, not easily extracted. In previous work screened out by the laboratory strain producing collagenase DL-12 is a strain of Serratia microbes from the ocean. In this experiment by UV mutagenesis approach to DL-12 addicted worm marcescens, Orientation screened strains yielding collagenase, to study its enzyme properties.DL-12 strain of UV mutagenesis treatment, the screening, skin screening by hydrolysis experiments transparent circle method to determine the high- yielding mutant strain collagenase.By screening out strains of enzyme activity compared to determine the best time of mutagenic 45 ~ 55 s. Elect enzyme activity than the original strain has significantly improved the three strains DL-12(50s)-3, DL-12(50s)-4 and DL-12(55s)-5.Then these three strains producing enzyme stability were measured and compared finalize mutagenesis strain DL-12(50s)-4 experimental species. Mutagenic strains DL-12(50s)-4 produce collagen protease activity significantly improved compared with the original strain, and stably inherited in its passages.For DL-12(50s)-4 mutant strain medium components and conditions were optimized. The results are: sucrose 4g; beef cream 10g; NaC l 5g; deionized water 1L; pH: 7.7. Under optimum conditions, the culture of collagenase activity was measured for 98.76 U / ml.Under optimal conditions for DL-12(50s)-4 extensive fermentation culture, the culture broth after 42 h was centrifuged, and dissolved with a phosphate buffer, placed in a dialysis bag and dialyzed to obtain a crude enzyme solution, enzyme activity can reached 483.9 U / ml.Collagenase has an important application of value. Mutagenesis strain DL-12(50s)-4 in culture, the collagenase secreted into extracellular therefore available in large quantities by liquid fermentation collagenase, while easily separated and purified. Now less productio n of collagenase producing bacteria source, mutagenesis bacteria DL-12(50s)-4 for the production of collagenase have great value.
