Amanita belongs to Amanitaceae of Agaricales of Hymenomycetes of Basidiomycota, and its species diversity is very rich, at present there is nearly400kinds reported in the world. Amanita toxin has some potential applications in cell biology, molecular biology, screening antibacterial, anti viral, anti tumor, etc. Vast majority of Amanita belongs to ectotrophic mycorrhiza fungi, symbiosis with plants under natural state, is a special large fungi. Its sustainable utilization will not be carried out because of Amanita resources rare, toxin value expensive and artificial domestication difficultIn Yunnan, Amanita are mainly symbiosis with yunnan Cyclobalanopsis glaucoides. C. glaucoides is an important representative of tree species from semi-humid evergreen broad-leaved forest in the plateau region of central region of Yunnan Province, and its distribution amount are large. To study on the biological relationship between Amanita and C. glaucoides is an important content on interpretation developmental mechanism of Amanita fruit bodies. But under natural environment it is difficult to study their symbiotic relationship using modern technology because of biological species complex around the mycorrhizal, and interference factors more. Indoor symbiotic system between Amanita and C. glaucoides must established in order to better study symbiosis mechanism. In this paper Amanita fritillaria and C.glaucoides as materials, C. glaucoides sterile system and pure symbiotic system-with Amanita fritillaria were preliminarily established.This provide a new direction or idea to enrich and perfect symbiosis scientific theory of ectotrophic mycorrhiza, to reveal Amanita mycorrhizal synthesis and fruiting body formation mechanism, to study Amanita artificial or half artificial domestication cultivation.The main research contents and results are as follows:1. C.glaucoides aseptic system established. The seeds of. C.glaucoides were taken as materials and germinate in axenic culture, callus induced by the stems and terminal buds of sterility plantlet. The results showed that suitable medium on callus induced by the stem section were MS+6-B A1.5mg/L+NAA0.05mg/L or MS+TDZ0.70mg/L+IAA1.0mg/L; suitable medium on callus induced by the terminal buds MS+6-BA1.5mg/L+NAA0.1mg/Lor MS+TDZ0.70mg/L+IAA1.5mg/L; callus proliferation medium MS+TDZ1.00mg/L+2,4-D0.3mg/L; root medium MS+NAA0.10mg/L while rooting experiment was done directly using seedlings that grew from terminal bud by which meantime callus inducted. 2.Pure co-cultivation of’Amanita fritillaria mycelia and C.glaucoides Callus. The optimization medium on Amanita fritillaria mycelia was10%green bean sprouts+PDA medium; mycelia and calluses were inoculated on co-cultivation medium(fungal medium optimized and calluses medium), mycelia grew best on co-cultivation medium3. outdoor symbiosis of Amanita fritillaria mycelia and C.glaucoides. plantlets.When plantlet’s root were inoculated Amanita fritillaria mycelia with4piece and5times finished, the plant height, chlorophyll content and SOD, CAT, POD, ATP enzyme activity of the seedlings were significantly higher than other treatments, showing the mycorrhiza may have been formed.
