Xiaoqing River is in the central Bohai sea drainage rivers of the Yellow River inShandong province. Xiaoqing River is the only hohai navigation, water and landtransportation channel, assuming the important task of Jinan bearing flood dischargeof the water in Shandong province.Within Xiaoqing River drainage’s jurisdictionareas,having developed industries and concentrated population, is an importantindustrial and agricultural production base in Shandong province. Therefore, it shouldbe paid much attention to the ecological environment on Xiaoqing River.In this thesis, the water quality and sediment bacteria groups were investigatedand a variety of flora analysis methods were carried on the comparison,in order tooffer an reference for keeping Xiaoqing River’s ecological environment andecological balance better,1.The water quality was analyzed according to the national standards.Some basicwater quality indexes were tested,such as the water temperature, pH value,conductivity,COD and so on.On the whole, the results showed that water quality wasgood.2.Plate count method was used to detect the total number of bacteria.Culturablebacteria analysis results showed that there were a variety of aquatic bacteria and someother soil bacteria. The Bacillus sp. was the most common bacteria.3.MPN method was used to detect the number of coliform bacteria, there was alarge number of coliform bacteria in individual locations.For example,4#(Huang)、7#(Ji)in Xiaoqing River(Jinan Reach)and4#(TKQ)、6#(LWZ)had alarger number of coliform bacteria. In these sites,sewage discharge phenomenon orother sources were likely to exist.4.MPN method was used to detect the number of nitrifying bacteria, nitritebacteria and denitrifying bacteria. The number of nitrifying bacteria was close to thenumber of nitrite bacteria. High number of denitrifying bacteria illustrated lowoxygen content in this site.5.The diversity of bacteria flora was analyzed according to the results from the16S rDNA sequence technologies of the partially isolated bacteria strains. Aquaticbacteria and some other kinds of soil bacteria existed in culturable bacteria, and the spore bacteria was the most common bacteria. In addition, some pathogenic bacteriawas divided such as mycobacterium.6.High-throughput sequencing technologies were used to detect the diversity ofbacteria flora. There was a high diversity of bacteria in sediment, but having a greatdifference.Xiaoqinghe river (Jinan Reach) drainage was mainly responsible for the outsideand sewage treatment, passing by the whole city water.The entrance on both sides ofthe wastewater was complex.It was of great significance to study the characteristic ofsediment microbial quantity for evaluating Xiaoqing river system and its environment.High throughput sequencing technologies could provide the outline of bacterialflora of sediment on the different taxonomic level, whereas hardly providing detailinformation of bacterial groups, such as the information of the bacterial at species.However, the traditional culturing technologies could provide the number of specificbacteria,such as coliform bacteria,nitrifying bacteria,denitrifying bacteria and somepathogenic bacteria. Each have its own advantages.Therefore,high throughputsequencing technologies combined with the traditional culturing methods could makeup for respective limitations, providing more comprehensive and detailed informationfor analyzing the microbial diversity in environmental samples.
