Traditionally, housekeeping genes refer to genes performing fundamental functions such as transcription or translation. Here, in this thesis, we use this term to refer to genes express constantly across different tissues. We use another term tissue enriched genes to refer to genes specific to only one kind of tissue. Based on microarray data, we obtained some housekeeping genes and tissue enriched genes in normal and cancerous tissues. After that, we did some comparison between these two kinds of housekeeping and tissue enriched genes. Using enrichment analysis, we found that housekeeping genes tend to enrich to fundamental functions and tissue enriched genes tend to enrich to tissue specific functions, which implies that our results are reliable. To further test the reliability of our results, we collected twenty-six datasets and picked out housekeeping genes not significantly differential expressed in all these twenty-six datasets. These genes may perform some most fundamental functions. We also did some network analysis on housekeeping genes and checked their roles in some diseases. Besides, we picked some disease susceptibility genes and did some enrichment analysis on the target genes of some transcription factors. |
