Phospholipid (PL) is an important part of cytomembrane, and it plays important parts in most activities of cells. As a kind of emulsifier and antioxidant, it is widely used in food, cosmetic and pharmaceutical industries. As an important source of co-3fatty acid, docosahexaenoic acid (DHA) plays important roles in many body functions. While having both functional properties of PL and DHA, phospholipid-docosahexaenoic acid (PL-DHA) also has many other unique biological activities, and is attracted much attention. Currently, food-grade PL-DHA is mostly exacted from marine organisms, but this kind of source has many limitations. The advent of industrial Upases and phospholipases provides more possible paths for preparation and production of PL-DHA.In this study, PL-DHA was prepared by two kinds of PLs used immobilized phospholipase Ai. Firstly, single factor method was utilized to optimize the conditions of acidolysis of phosphatidylcholine (PC)-rich PL with DHA, using the content of DHA in product as standard. The influences of different parameters, such as molar ratio of substrates, reaction temperature, enzyme dosage, solvent dosage, and water activity, were investigated systematically. Then the response surface method was used to optimize the conditions continuously. The content of DHA was17.5mg/g PL under the optimal conditions.Secondly, single factor method was utilized to optimize the conditions of acidolysis of high-pure soybean power phosphatide with DHA, using the content of DHA in product as standard. The influences of different parameters, such as molar ratio of substrates, reaction temperature, enzyme dosage, solvent dosage, and reaction time, were investigated systematically. Then the response surface method was used to optimize the conditions continuously. The content of DHA was70.1mg/g PL under the optimal conditions.Finally, according to some data in the optimization, the mechanism of acidolysis of PL with DHA was discussed simply.
