D-7-ACA(3-deacetyl-7-Aminocephalosporanic acid),a new intermediate of cephalosporin antibiotics,it has a lot of advantages comparing with 7-ACA (7-Aminocephalosporanic acid).Using the intermediate the cephalosporin antibiotics is simpler in producing,easier to be modified and purified,better in quality and less in cost etc.The technology is completed by the tri-enzyme method,using cephalosporin C as materials to prepare D-7-ACA through after-treatments.Preparing D-7-ACA with tri-enzyme method(Immobilized D-amino acid oxidase,Immobilized GL-7-ACA Acylase,Immobilized deacetyl esterase) is an original,sustainable and brand new technology in promoting the development of green pharmaceutical industry in our country.D-7-ACA is highly efficient in preparing cephalosporin antibiotics,namely,cefuroxime, cefoxitin,cefpodoxi(?)e proxetil,etc.This paper has found the best way to prepare D-7-ACA through changing the concentration of substrate,PH value and temperature applied in the reaction process.The tri-enzyme used in this technology promotes the conversion to 92% with a final yield of 82%.The optimized enzymetic technology is using cephalosporin C(Fermentation extract) as material to produce D-7-ACA with its yield achieving 82%.As a result,it makes full use of the fermentation extract and we have extablished an effective analysis way.
