ATP is very important in all living beings as a carrier molecule for energy, and many enzymatic reactions proceed simultaneously with hydrolysis of ATP in the biosynthetic reactions. Therefore, ATP is widely used in medicine to activate metabolism of the brain, blood vessels, etc. Recently, ATP is used to cause both necrosis and apoptosis and is effective against cancer.In this dissertation, ATP production from adenine by Actinomucor elegans had been studied systematically, including the optimization of Actinomucor elegans culture conditions by response surface methodology, study on the basic reactive conditions for ATP production by Actinomucor elegans, effects of cell permeability on the activity of Actinomucor elegans for ATP production from adenine, effects of supplementary material on the activity of Actinomucor elegans for ATP production from adenine.Response surface methodology (RSM) was used to optimize the culture conditions for ATP production by Actinomucor elegans. The results showed that the optimum conditions were as follows:temperature 33℃, agitation160r/min, time10h, glucose51.54g/L, yeast extract5.22g/L, corn steep liquor14.31g/L, MnSO4·7H2O0.5g/L, FeSO4·7H2O0.1g/L, NH4C13g/L, K2HPO4·3H2O3g/L, pH6.0~6.5, which increased the productivity of Actinomucor elegans from23.51g/L to31.13g/L, the percent conversion of adenine from53.59%to59.97%and the productivity of ATP from6.56g/L to7.34g/L.The basic reactive conditions for ATP production from adenine by Actinomucor elegans were investigated comprehensively. Based on shaking flask experiments with orthogonal experiment, the optimum reaction liquid was obtained. After optimization in production process,8.51g/L ATP was accumulated from3g/L adenine in12h and the molar conversion ratio was69.53%, which increased by41.83%compared with the control group.Methods of physics and chemistry related to the modification of the cell permeability and cell enzymatic activity for ATP production were studied in Actinomucor elegans. The correlation between cell membrane permeability determined by the electrical conductivity method and the cell enzymatic activity for ATP production was studied. As a result, drying method was the best way to increase the cell enzymatic activity for ATP production by Actinomucor elegans. After optimization in production process,10.13g/L ATP was accumulated from3g/L adenine in12h and the molar conversion ratio was82.76%, which increased by 202.39%compared with the control group.Effects of supplementary material on the activity of ATP production from adenine were studied preliminarily in Actinomucor elegans. After optimization in production process,18.33g/L ATP was accumulated from8g/L adenine in14h and the molar conversion ratio was56.07%, which increased by179.42%compared with the control group.
