Ubiquitin-proteasome pathway (UPP) is a pathway obtained mediated proteindegradation by ubiquitin, plays an important role in the maintenance of cell function.Research shows that, the protein ubiquitination degradation pathway related to theoperation of cell cycle, and the development of the embryo, and also plays animportant role in hormone and aging. Protein ubiquitination activated the essence ofprotein to be degraded, the process is completed with the participation of a number ofenzymes, including ubiquitin activating enzyme E1, ubiquitin conjugating enzyme E2and the ubiquitin protein ligase E3. Ubiquitin molecules will target proteinubiquitination in the role of these three enzymes, ubiquitin protein26S was finallyproteasomal recognition and degradation. Ubiquitin conjugating enzyme E2has animportant role by the ubiquitin proteasome pathway in plant. The previous work inour laboratory has shown that, in the wheat leaf rust interaction, whether the level oftranscription or translation level, E2gene were upregulated in early defense responseof incompatible interactions, preliminary analysis of its resistance to leaf rust infestedwith defensive reactions are closely related in wheat. In order to further study thefunction of TaE2, the test is based on. Gao Xiang has been successfully constructedthe eukaryotic expression vector pSuper1300+-TaE2, and transformed intoArabidopsis thaliana obtained by over expression of TaE2gene in T1generationplants Agrobacterium; at the same time, purchased from the SALK Library ofArabidopsis thaliana T-DNA insertion mutants were obtained. Molecularidentification of Arabidopsis AtE2gene mutation in homozygous (AtE2). The test iscontinued on the basis of the screened separation does not occur in the ratio of T3generation of over-expression TaE2homozygote (OE), and Agrobacterium Ate2mutants obtained TaE2revertant plants homozygous positive (Ate2::TaE2). On thebasis of four genotypes of the obtained Arabidopsis thaliana (wild type, AtE2,Ate2::TaE2, OE) conducted abiotic and biotic stress, in order to reveal the wheat E2gene in plant resistance to environment stress effect. Test also use wheat matureembryo transformation system,of Agrobacterium mediated transformation,Agrobacterium transformation of wheat in order to obtain enhanced resilienceoverexpression TaE2wheat lines.The main results were shown as follows: 1We obtained the overexpression of Arabidopsis TaE2gene positivehomozygote plants.2We obtained the pSuper1300+-TaE2mutation positive homozygote plants.3By salt, drought, poor nutrition and other abiotic stress test, Our studyconfirmed that the TaE2gene has an important role in plant resistance under abioticstress.4By infecting arabidopsis with powdery mildew we confirmed that TaE2geneplays a positive regulation role in the process of arabidopsis and powdery mildewinteraction.5By construction the pCAMBIA-TaE2enpression vertor and use the wheatembryo transformation system by Agrobacterium mediated transformation Zhengzhou5389,obtained generation plants of T1.6The generation plants of T1wheat plants were inoculated with leaf rust race165and FB staining, the results showed that after96h the OE plants mycelia is small thanCK. Preliminary indication of expression TaE2inhibitory effect on mycelialgrowth,indicating TaE2has resistance in wheat and rust interactionresistant process.
