Zinc ions participate in a variety of metabolic processes in human body, such as neurotransmitter and the catalytic activity of enzymes and cell apoptosis. Deficiency of zinc in human body can cause metabolic disorders, resulting in a variety of neurological diseases such as Alzheimer’s disease and Parkinson’s disease. The content of zinc in the soil is vital for the growth of plants, In the field of industry, the production and use of zinc can also produce a large amount of waste water containing zinc, causing certain harm to human body and environment.This research developed a new series of fluorescent probe using biological method to detect the contents of Zn2+, which is based on CFP-PML-CFP. Using FRET technology on zinc finger protein PML and its related proteins, we constructed a series of new type of Zn2+fluorescent probes, namely Zifl, Zif2, CFP-PML-PML-YFP, CFP-RBB-YFP, CFP-PML and PML-YFP. Among them, the detection upper limit of the mutant types Zifl and Zif2to zinc ions is10μM. CFP-PML-PML-YFP fluorescent probe’s detection upper limit is40μM. CFP-PML and PML-YFP’s detection upper limit is80μM. However, the detection range of CFP-RBB-YFP to zinc ions can be as high as1-1000uM. The fineness of detection of these probes to zinc ions is Zifl, Zif2> CFP-PML-PML-YFP>CFP-PML, YFP-PML> CFP-RBB-YFP. Through the analysis and comparison to these probes, we obtained a series of specific and efficient fluorescent probes of different detection upper limit, which can quantitatively detect Zn2+in living organisms and the environment.
