With the more and more serious global warming trend, the harm of high temperature on crop has increasingly received people’s attention. Rice as a kind of native to tropical and subtropical thermophilic crops, its every stage has certain appropriate temperature range. The temperature which exceeds the range is not conducive to the growth and development and will cause serious harm on rice growth, yield and quality formation.MicroRNA belongs to the smallest functions ncRNA of animals and plants, which can combin with specific complementary loci on target mRNA and inhibit the expression of genes in the transcription level. Some microRNA shows different developmental stages or strict specificity model expression in different tissue cells and tissue types, while other miRNA expressed widely in the organisms. Related studies have shown that microRNA is involved in model plant arabidopsis floral organ development and flowering regulation process, and temperature effects on rice flowering earing period is also very important. So the research on high temperature stress on rice also becomes an urgent need.In order to find out the microRNAs which have different expression, the research compares the different expressions of miRNAs in rice earing flowering period with appliying the Proton platform analysis, and to verify the significant microRNAs expression differences in more samples through the qRT-PCR method.(1)Collect rice flowers which include the flower of the control group and the mixture groups after high temperature stress, and then analysis the miRNA expression differences with the application of Ion Proton platform (genomics company). The whole process includes the RNA extraction from tissue samples, library construction, titration and sequencing in the sequencing platforms and analysis using the relevant data analysis software.. Finally,23 differentially expressed microRNAs were gotted.(2) Total RNAs were extracted from the 6 frozen rice flower samples after high temperature stress, and then reverse transcription into cDNA for subsequent experiments. Primers designed according to the miRNA database and related literature. And verify the expression differences of these miRNA which were under the high temperature stress in flowering period, including Osa-miR159c、Osa-miR159d、Osa-miR159f Osa-miR156c-3p、Osa-miR167c-3p、Osa-miR164d、Osa-miR529b、Osa-miR528-5p Osa-miR166b-3p、Osa-miR166h-3p.
