Recent studies show that some guanine-rich oligonucleotides with GC termini are able to form prolonged G-quadruplex-based superstructures termed G-wires.G-quadruplex is a special kind of second level structure, they are likely to rebuilt in the G-rich solutions. The construction of specific surface architectures via controlled self-assembly is a key goal in the design of nanoscale molecular electronic devices.The formation of G-quadruplexes can make the structure of genome stable,is a useful investigation of against cancer and anti-decrepit,also has very important meaningful significance.From 2013 people find that G-quadruplexes are naturally existed in human cancer cells, this important research makes people are highly concentrate on the study of G-quadruplexes.Since it has been proposed, people research it in system quickly. What a different is that we use higher molarity of oligonucleotides with GC-terminal ends samples than before.Compare to the samples with the different GC-ends situation have the different results and new searching.In this work, we report on the sequence with GC-end and another sample without GC-end,they are in the same situation of Na solvent. Using dynamic light scattering, ultraviolet-visible spectroscopy, nuclear magnetic resonance doing the measurements to research the formation of G-quadruplexes and G-wire grows. We have four chapters, mainly showed as follow:The first chapter represent that fundamental components of DNA and G-quadruplexes, the background introduction to G-quadruplexes. And the progress of G-quadruplexes investigation around the world. Supposed to the optical method can be used very well. Showing the significance of this study.The chapter two introduces the basic theory of the optical method which are fully used in our research. First of all, the characteristics of dynamic light scattering theory,also give the mathematical expressions of this theory. How to measure the samples with dynamic light scattering setup. Secondly, with ultraviolet-visible spectroscopy measurements to verify if G-quadruplexes occured in solution. Then give the simple introduction of UV-vis theory. The last, we described how the NMR experiments works with our typical samplesThe chapter three are interestingly interpreted specific operate measurements. First the preparation of sample solutions, to calculate the concentration of samples and how to made them. Structure diagram have been illustrated in this paper. When we got the parameters of our measurements, we use the origin to fitting the curves. Then according to the theory of the measurements analyse all the data, give a final reasonable conclusion.Chapter four, the results and conclusion of our measurements, discussions. Interpreted there are G-quadruplexes and G-wire grows in our samples. On the basis of the foregoing paper made a summary and expectation.
