Bta-miR-21-5p is located on chromosome 19 in bovine genome. The expression of bta-miR-21-5p and interaction with the target genes affect cell growth, migration and apoptosis. The present study was designed to investigate the expression and its target genes of bta-miR-21-5p in fertilized, nuclear transfer cloning and transgenic bovine fetus fibroblast cells. Three target genes of bta-miR-21-5p were sequenced and predicted, which are fibroblast growth factor-10 (FGF-10), lipoprotein lipase (LPL) and adipose triglyceride lipase (ATGL). After transfection of bta-miR-21-5p mimic into bovine fetal fibroblast cells and 3T3-L1 fat cells, respectively, bta-miR-21-5p expressed increased in both fetal fibroblast cells and 3T3-L1 cells, while the target gene FGF-10 and ATGL expressed decreased. After transfection of bta-miR-21-5p inhibitor, the bta-miR-21-5p expressed decreased, and FGF-10 and ATGL expression increased. These results suggested that bta-miR-21-5p and FGF-10 and ATGL was negatively correlated. The LPL expression was with no obvious correlation with bta-miR-21-5p. These results suggested that bta-miR-21 probably negatively control the target genes FGF-10 and ATGL. |
