The plants always suffer stress that come from environment in its process of growth and development,when the plants suffer stess, the concentration of calcium ions in thecytoplasm occurs cyclical shocks and form "calcium signal". The transmission of stress signal is carried out by the calcium binding protein. The calcium binding protein has been reported mainly include CAM, CDPK, CBL and other ones. CBL is an unique calcium binding protein of plant that found in Arabidopsis thaliana L.. CBL and its downstreamtarget gene family CIPKs play a role. CIPK is a kind of serine/threonine protein kinases family which special existence in plants. CBL-CIPK signaling network system research in model plant arabidopsis and rice is detailed and deeply, however, the study in Sinapsis alba L. has no reported. Sequencing of the genome sequence of arabidopsis thaliana provide an important basis and reference for the study of the homologous Sinapsis alba.L’s CBL-CIPK stress answering network system. Our lab carried out cloning and functional study of CBLs gene family members of Sinapsis alba L. for the first time, and achieved a certain degree of accumulation, the research focuses on CBL3 of Sinapsis alba L.CBLs family members, the main results are as follows:1) Through the method of homologous cloning, we cloned CBL1、CBL 2、CBL 3、CBL 4、CBL 9、CBL 10 gene from Sinapsis alba L.,the length of them are 276 bp, 291 bp, 305 bp, 188 bp, 435 bp and 384 bp.2) By the technology of RACE,we got the whole length of CBL3,the CDS gene sequence length of CBL3 is 681 bp, encoding 226 amino acid residues, its molecular weight is 25795.3, the instability index is computed to be 39.65, grand average of hydropathicity(GRAVY) is-0.204.sequence homology and phylogenetic tree analysis, this gene and Arabidopsis thaliana CBL3 gene are homology, so we can named it Si CBL3.3) Using PEG6000 deal with Sinapsis alba L..seedings and test the expression of Si CBL3 in different time with real-time PCR,we found Si CBL3 expression of the leaves increased in the early stage of drought stress, with the extended treatment timeof drought stress, the expression of Si CBL3 showed an decline trends,what illustrates Si CBL3 gene in plant response to drought stress and prevent plant from further damage.
