In this dissertation,the femtosecond laser cell micromanipulation system is studied. The main contents are classified as follows:1. The femtosecond laser cell micromanipulation system was realized. A key part of the system is a commercial inverted research microscope (IX71, Olympus). Femtosecond laser pulses were introduced into the microscope through an image path from the left side port. We designed motorized specimen stage with a resolution of 0.2μm and modified the image parts. Based on these grounds, an experimental instrument of femtosecond laser cell micromanipulation system was built.2. Stable optical trapping of human red blood cells (RBCs), Haematococcus pluvialis Flotow cells and Xanthophyllomyces dendrorhous cells was achieved using this system.3. We measured the optical trapping capability and Q-values of the femtosecond laser tweezers in comparison with CW laser tweezers. In the experiments, we found that this two kinds of laser tweezers exhibited different optical trapping capabilities.4. The influence on the activity of cells captured by CW laser tweezers and femtosecond laser tweezers was studied using staining method.5. A homemade micromanipulator was built and the isolation of single cells was performed.6. A detection system of light scattered by samples was installed on the inverted research microscope and a pump-probe system was built outside the microscope, which is the groundwork of studying on the ultrafast biologic process.7. At last, we make some suggestions of the future work, which gives a direction for further study on femtosecond laser cell micromanipulation system.
