Polymerase Chain Reaction Microchip (PCR Microchip) appeared at the beginning of the 1990s. It is fabricated by the MEMS technology on the substrate of silicon, glass, polymer etc. There is a series of micro channel, micro chamber, micro heater, and sensor etc in PCR microchip. In contrast with traditional PCR thermocycler, PCR microchip has some advantages, such as small size, high efficiency.Temperature control is crucial to PCR microchip. In this dissertation, heat conduction mechanism for PCR microchip is explored, and mathematical model is studied. On this basis, the numerical simulation is made for combined transient heat transfer with natural convection and thermal radiation using CFDRC software. The efficiency of heating and cooling of microchip is analyzed theoretically.The topics of this dissertation are presented below:1. On the basis of the work in our laboratory, heat conduction mechanism for PCR microchip is explored, and mathematical model is studied;2. CFDRC software is studied thoroughly, and user defined function is written because of non-constant heating power of the microchip and pulse heating. Heating, cooling rates and temperature distribution to the microchip are analyzed using numerical simulation method. The analysis results prove that the microchip has higher temperature rising and temperature distribution of the sample can meet the requirement of the PCR.3. Heating and cooling rates are solved in theoretical way by simplifying rationally heat transfer of the microchip. And the simulation results of the simplified model are almost identical with theoretical solution.The dissertation showed that numerical simulation method has some advantages suchas convenience and accuracy, as well as breaking through the restriction of the experiment method. It is believed that numerical simulation method will be adopted in a large amount in the Micro Total Analysis Systems (uTAS) research, and promote the development of uTAS research.
