Objective:to explore the association of EGF gene with gout inflammation.Methods:(1)Respectively, using 25,50 and 75 ug/ml MSU crystals stimulate the human monocyte cell line(THP-1) which were treated with 0.5?M PMA for 3 hours. The expression level of EGF and IL-1βgenes were detected by Quantitative Real-time PCR. The content of IL-1β in the supernatant was measured using an ELISA kit. The differences of genes expression level and IL-1β content in the supernatant were analyzed between multiple groups.(2)Peripheral blood mononuclear cells(PBMCs)of 76 control subjects,58 patients with acute gout and 122 patients with intercritical gout were collected. The level of EGF m RNA was also detected by Quantitative Real-time PCR. One-way ANOVA was used to analyze data between these three groups.Results:(1)the expression of IL-1β m RNA in 25,50 and 75ug/ml MSU groups were significantly higher than the group only treated with PMA(P<0.05,respectively). Futhermore,the content of IL-1β in the supernatant of 25,50 and 75ug/ml MSU groups were also significantly higher than PMA group(P<0.05,respectively). But the expression of EGF m RNA between these four groups have no significant difference(F=1.961,P>0.05).(2)The expression of EGF m RNA in patients with acute gout and intercritical gout both increased significantly compared to healthy controls(P<0.05,respectively). Moreover, Significant difference was also observed between patients with acute gout and intercritical gout(P<0.05).Conclusion: EGF is closely associated with gout inflammation, and may play a role in regulating inflammation of gout.
