Goodyera schlechtendaliana Rchb.f., a member of the genus Goodyera, is an orchidaceous perennial herbaceous plant. Its whole herb has been widely used with moistening lung cough, nourishing kidney, enriching qi, promoting flow of qi and blood circulation, and swelling detoxification effects. The plant is used for the treatment of tracheitis, dizziness and fatigue, and neurasthenia, it can also be used for external application to treat breast carbuncle, furuncle, poisonous snakebites and scrofula, and the Chinese pharmacopoeia has not contained. G. schlechtendaliana has a long history of folk application and definite therapeutic effects, but few researches on its chemical constituents, pharmacologic actions, pharmaceutic preparation and quality control have been reported, further exploitation and utilization is limited. In this experiment, carried out G schlechtendaliana chemical composition analysis and its application, the specific contents and results are as follows:1. Chemical extraction and structure identification of G. schlechtendalianaIn this paper, petroleum ether and ethyl acetate parts of 95% ethanol extract were separated and purified through silica gel, ODS and Sephadex LH-20 column chromatography and semi preparative HPLC, seven compounds were obtained and their structures were identified as syringaldehyde (I),5-hydroxymethylfurfural(II), alloimperatorin(III), vanillic acid(IV), ferulic acid(V), glyceroyl monopalmitate(VI) and β-sitosterol(Ⅶ) by’H-NMR、13C-NMR、MS data and comparision with those in literature. Compounds II-VD were isolated from this genus and compounds Ⅰ-Ⅵ were isolated from this plant for the first time.2. Antioxidant activity of G. schlechtendaliana extractionThe in vitro antioxidant activities of petroleum ether, ethyl acetate and n-butanol parts, and the main compounds were measured through DPPH and potassium ferricyanide reduction methods,the clearance of DPPH was used to test the scavenging ability,and the total flavonoid and polyphenol contents were determined. The results showed that the DPPH was inhibited by FA, the scavenging ability of FA was equvivalent to the scavenging ability of vitamin C. Ethyl acetate part was significantly higher than petroleum ether, n-butanol parts significantly. The reducing power of FA and VA were higher than three extract parts significantly. The ethyl acetate part of G. schlechtendaliana has the most strongest scavenging ability of three extract parts, the strongest antioxidant activity and the highest total flavonoids and polyphenol contents,higher than petroleum ether, H-butanol parts significantly (p<0.05,p<0.01). Orthogonal experiment was used to optimize the extraction process of G. schlechtendaliana with ferulic acid and the extraction yield of ethanol as evaluation index. The optimal extraction process was as follows: the concentration of ethanol was 70%, the ratio of sample to extract solution was 12:1, the extract time was three.The percent of extractum was 14.81,one gram of extract appropriate to 6.75 gram of G. schlechtendaliana..3. Cream preparation of Goodyera schlechtendalianaThe ethyl acetate part of G. schlechtendaliana cream was prepa red based on the above optimal extraction process; the preliminary study of quality control was carried out; appearance, particle size, pH value, potential and the main component content were involved. The results showed that:O/W cream was mooth and easy to coat; the average particle size of dispersed phase was 84.48 nm, potential, was 41.6 mV, pH value was 6.76; G. schlechtendaliana cream, ferulic acid and vanillic acid appeared in the same position by TLC analyses; the contents of ferulic acid and vanillic acid were 0.152% and 0.037%, respectively.The preliminary study on composition, antioxidant activity, extraction process, cream preparation and quality control of G schlechtendaliana have been carried out in this paper, the results provided certain theoretical basis in developing it into cream preparation.
