Purpose: To discuss Danqing granule’s curative effect about intrahepatic cholestasis of rats with heat syndrome induced by ANIT,analyze and probe it’s mechanism.Material and method: 60 healthy and clean Wistar rats were randomly divided into blank control group 10 and model group 50(Model group 1, model group 2, Yinzhihuang granule group, Danqing granule low-dose group, Danqing granule high-dose group, 10 each group).Rats in the blank control group were gavaged by 0.9%NS, the remainder rats in model group were placed the heat modeling box, and mixed gavaged by 50% glucose and pig fat every day for consecutive 3 weeks to analog TCM heat syndrome. The 22 nd day, the 50 rats in model group were gavaged by 2%ANIT salt solution disposablly, copy the acute intrahepatic cholestasis in rats model with heat syndrome. Model group 1 was putted to death after modeling 24 hours, serum specimens, bile and other specimen to spare observe liver tissue sections under the Light microscopy, to make the model successfully. Model group 2 was gavaged by 0.9%Na Cl every day, from the modeling day; The remaining rats in intervention group were gavaged by corresponding drugs, for consecutive 7 days. The 29 th day, all the rats were putted to death, and collected specimens. Observe different effects of drugs about the liver function, bile composition, liver lipoprotein X of model rats, obverse the pathological changes of liver under the Light microscopy, obverse the ultrastructure of liver under the Electron microscopy.Results: 1.Compared with blank group, the rats in two model groups, its’ physiological state were less well; each serum biochemical indices(ALT,AST,ALP,TBIL,DBIL and TBA) and lipoprotein X increased significantly(P<0.05), the bile flow reduced, and velocity of flow slowed(P<0.05), the LEC and TC of bile reduced, the TBA and TB of bile increased, the differences all have statistically significant(P<0.05). We can see the liver cell swelled, level arrangement disordered. and a lot of inflammatory cells and fat cells under the Lightmicroscopy; Mitochondrial in cytoplasmic were swelled and ruptured, changed like vacuolar under the electron microscopy. 2.Compared with model group 1 and 2, the level of ALT,AST,ALP,TBIL,DBIL and TBA of serum and lipoprotein X all reduced(P<0.05),the degree of liver tissue’s damage is approaching under microscopy. 3.The rats in three therapy groups compared with model 2, general state changed better,every serum indices and lipoprotein were decreased obviously(P<0.05).The bile flow increased and velocity of flow accelerated(P<0.05). The LEC and TC of bile increased, the TBA and TB reduced. All differences had statistically significant( P<0.05).The liver tissue and ultrastructural changed better under microscopy. 4.Compared with Yinzhihuang group, the rats in Danqing granule high-dose and low-dose group had better general state. The level of TBIL,DBIL and TBA of serum reduced obviously(P<0.05). The liver tissue and ultrastructural restored better under microscopy. 5.Compared Danqing granule high-dose and low-dose group, every experimental index’s difference is not significant(P>0.05), the liver tissue and ultrastructural were same basically under microscopy(P>0.05).Conclusion: 1.Danqing granule can improve the physiological state of intrahepatic cholestasis rats with heat syndrome. 2.Danqing granule can improve liver function, promote the secretion of bile excretion and reduce the liver cell lesions and necrosis. 3.Danqing granule has a positive therapeutic effect in intrahepatic cholestasis with heat syndrome.
