Effects Of Acupuncture Combined With Yiqi Qufeng Decoction On Inflammatory Cytokines IL - 1β And TNF - α In ACD Mice

Purpose:Allergic contact dermatitis, eczema and other skin diseases is associated with type Ⅳallergy disease, appears in local redness, rash,itching, symptoms such as chickenpox acute or chronic dermatitis. This experiment by using 2 nitro fluorine benzene(DNFB) to establish allergic contact dermatitis in mice, by detecting the thickness, ear weight, ear ear pathology and cytokines, to observe the traditional Chinese medicine yiqi functions of soup in combination with the effects of acupuncture on ACD histopathologic of mice, and to explore its action mechanism.This experiment by using 2 nitro fluorine benzene(DNFB) to establish allergic contact dermatitis in mice, by detecting the thickness, ear weight, ear pathology and cytokines, to observe the traditional Chinese medicine,The soup of Yi Qi Qu Feng in combination with the effects of acupuncture on ACD histopathologic of mice, and to explore its action mechanism.Material and method:1.60 kunming mice were randomly divided into 6 groups(20±2) g, namely blank control group, model group, positive drug group(loratadine), acupuncture, Chinese traditional medicine group, the combination of acupuncture with medicine group.2.In the experiment on the first day(sensitization) back shaving area and with 7% DNCB(mixture solvent of acetone: olive oil = 4:1) 25 mμL sensitization, repeat 1 times the next day. After 5 days(period) induced mice ear inside and outside with 0.5% DNCB5 mμL, inducing ACD. Blank group mice also same amount of distilled water. Each infusion medicine since before 1 d sensitization, 1 times a day, 7 d in a row. In the last 6 h after excitation to medicine. Observation group after mice induced dermatitis ear thickness difference, poor quality of ear, ear pathology morphology. Using immunohistochemical method(Immunohistichemical method) to determine the type of serum interleukin 1 beta(IL- 1 beta) and tumor necrosis factor alpha(TNF alpha) expression and observe the influence of gas on cytokine secretion functions of soup;Application of HE staining to smear of pathology, in order to observe the soup of Yi Qi Qu Feng on the effects of ACD diseased tissue pathological changes of mice; By comparing each group and the difference of thickness difference, poor quality of ear in mice, the evaluation of combination of acupuncture with medicine on the therapeutic effect of allergic contact dermatitis and explore some of its action mechanism.Results:1.The soup of YiQiQuFeng can effectively reduce the pathological changes in mice ear thickness difference and ear weight difference, inhibition of ACD inflammation pathological changes of mice.(P<0.05)。2.Compared with model group, acupuncture group, yiqi functions of soup, the combination of acupuncture with medicine group, loratadine group can cut the content of serum TNF alpha ACD mice. Between groups had significant difference compared with ACD model group(P < 0.05), the combination of acupuncture with medicine groups with loratadine group’s influence on the level of serum TNF alpha, no obvious difference(P > 0.05), the combination of acupuncture with medicine group compared with other treatment group have difference(P < 0.05);3. Acupuncture group, yiqi functions of soup, loratadine group, the combination of acupuncture with medicine group can cut ACD diseased tissue in mice in the expression of IL- 1 beta, and ACD model group compared with significant difference(P < 0.05); Loratadine group and the combination of acupuncture with medicine on ACD diseased tissue of mice in the expression of IL- 1 beta had no significant difference(P > 0.05), the combination of acupuncture with medicine group compared with other treatment group have difference(P < 0.05).4. The ACD model group mice diseased tissue pathological slices showed epidermis and the dermis has obvious inflammatory cell infiltration, tissue interstitial edema.The soup of Yi Qi Qu Feng after the intervention, the skin epidermis and the dermis, inflammatory cells infiltration in significantly reduced tissue interstitial edema, reduce obviously.Conclusion:1.The soup of Yi Qi Qu Feng can effectively reduce the pathological changes in mice ear thickness difference and ear weight difference, inhibition of ACD inflammation pathological changes of mice.2.Acupuncture combined with the soup of Yi Qi Qu Feng can decrease the IL- 1beta in the diseased tissue in mice, and the expression of TNF alpha.3.The results showed that acupuncture combined with allergic contact dermatitis has certain inhibition of inflammation, its mechanism may be related to combination of acupuncture with medicine can regulate the body’s immunity, inhibit Th1 lymphocytes activation to reduce certain inflammatory cytokine secretion, regulating Th1 / Th2 immune imbalance, restrain skin inflammatory reaction.4.Acupuncture combined with Chinese traditional medicine in inhibiting allergic contact dermatitis(inflammation of some aspects can be achieved and the effect of glucocorticoid hormones.
