Gastric cancer is one of the most common digestive tract malignant tumor in the world,the surgery is the main treatment of gastric cancer,but after resection, there are still a part of the patient’s tumor recurrence and metastasis happens.Research think that leads to tumor recurrence is cancer stem cells. However surface specific markers to identify cancer stem cells,and by studying the biology and pathology characteristics determine their role in tumor development and prognosis.In this study, through the screening of stem cell marker,Identify whether expression in gastric cancer cells.By indirect fluorescent antibody technic double-staining,to detect the expression of CD133+/CD14+ in gastric tissues,Identification gastric cancer stem cells and to analyze its clinical significance. By primary gastric tissue for cell culture after, through the Real-time Quantitative PCR Detecting(Q-PCR), comparative gastric cancer tissue and tissue adjacent to carcinoma of CD133 gene and CD14 gene expression. Meantime to investigate the effect of arsenic trioxide(As2O3) on the proliferation and apoptosis of gastric cancer cells NCI-N87.The cell proliferation was detected by MTT, and cell apoptosis was detected by TUNEL.Experiments show that CD14 positive in gastric cancer cells. CD133 + / CD14 +is markers for gastric cancer identification, can also be used as important reference index of early diagnosis and prognosis judgement,and the gastric cancer tissues CD133 gene and CD14 gene expression were higher than tissue adjacent to carcinoma.As2O3 can promote the apoptosis of gastric cancer cells, which can inhibit the proliferation of gastric cancer cells NCI-N87.The result of this experiment for gastric cancer stem cells added new markers, and the treatment and prognosis of gastric cancer provide important experimental basis, also for gastric cancer cell proliferation inhibition provides a new idea.
