Depression after acute myocardial infarction is clinical common complications after myocardial infarction, on the one hand, after myocardial infarction patients increased the risk of depression, however, the depression may increase myocardial infarction in patients with symptoms after myocardial infarction either. Tranditional Chinesemedicinehave already associated with heart and brain. They think that the relationship between the heart and brain are inseparable. Modern medicine in the treatment of depression after myocardial infarction is still using antidepressants, rather than the combination of heart and brain. Modern medicine has found that the correlation of heart and brain, but its biological mechanism is not yet clear. We collecting the reference and data to further prove that heart and brain associated biological mechanisms. Provide reliable basis for clinical treatment and theories, rich in heart and brain correlation theoryObjective:This study aimed to investigate both the behavior and morphology changes of rats after acute myocardial infarction in order to provide animal models for studying the mechanism of depression after myocardial infarction. This study aimed to evaluate the behavior,brain issue HE training and the expression regulation of the inflammatory such as TNF-a, IL-6 and apoptosis such as Caspase-3 and Caspase-8. Evaluation the relationship of inflammatory and depression behavior, apoptosis and depression behavior, inflammatory and apoptosis in rat brain tissue. To investigate the morphology changes of rats after myocardial infarction, and to investigate the treatment with pentoxifylline. To reveal occurrence mechanism of the depression after acute myocardial infarction by judging the depression symptom and detecting the protein content of TNF-a, IL-6, Caspase-3 and Caspase-8.Establishment of the mechanism relationship between Inflammatory factor、 apoptosis and the depression behavior after acute myocardial infarction. To reveal the occurrence mechanism about inflammatory and apoptosis after acute myocardial infarction, providing the drug target in depression after acute myocardial in clinical drug treatment. Methods:Cotton thread was observed in acute myocardial infarction operation. Rat randomly divided three groups:Sham group, Model group and PTX group. To observe the EGG for each group before acute myocardial infarction operation and after acute myocardial infarction operation in 20 minutes. PTX, inflammatory inhibitor, was given with intraperitoneal injection to the PTX group. Sucrose water consumption test, open-field test and forced swimming test were used to evaluate the behavior changes of the rats after acute myocardial infarction model operation. And observation of rat brain tissue HE staining. Meanwhile, the detection of TNF-α and IL-6 in both serum and brain of rats were evaluated to figure out the changes of inflammatory cytokines after acute myocardial infarction. At the same time, the detection of Caspase-3 and Caspase-8 in brain of rats were evaluated to figure out the level changes of apoptosis. Enzyme-linked immunosorbent assay (ELISA) were used to detect the level of TNF-α and IL-6. Western blot were used to detect the level of Caspase-8 and Caspase-3.Bivariate related and simple linear regression analysis were used to evaluate relationship of inflammatory and sucrose water consumption, apoptosis and sucrose water consumption, inflammatory and apoptosis in each group rat brain tissue (Sham group,Model group, PTX group).Results:The EGG show the ST section improved significantly after rat acute myocardial infarction. After two weeks rat acute myocardial infarction, compared with the sham group, the sucrose water consumption test of model group were significantly decreased(P<0.01), rat were shown the lower interesting in sugar water. Compared with the sham group,the totally movement and the totally movement time and stand up times of model group in open filed test were significantly decreased(P<0.01), and the swimming time and struggling time of model group in force swimming test were significantly decreased,meanwhile,the immobility time in model group were significant increased(P<0.01). Compared with the sham group, the quantity of inflammatory cytokines TNF-α, IL-6 in both serum and brain of model group were significantly increased(P<0.01), the protein levels of apoptosis Caspase-3 and Caspase-8 of model group were significantly increased(P<0.01).Compared with the sham group, the inflammatory cytokines in the brain issue HE staining of modal group were significantly increased and showing brain issue significantly disorder of model group. However, Compared with the model group, the sucrose water consumption test of PTX group were significantly increased(P<0.01),the totally movement and the totally movement time and stand up times of PTX group in open field test were increased(P<0.05 or P<0.01),the swimming time and struggling time of PTX group in force swimming test were increased,meanwhile,the immobility time in PTX group were significant decreased(P<0.01). Compare with the model group and PTX group, the bivariate related of TNF-α and sucrose water consumption, IL-6 and sucrose water consumption, Caspase-3 and sucrose water consumption have no relationship (P>0.05) in Sham group. And the bivariate related of TNF-α and sucrose water consumption, IL-6 and sucrose water consumption, Caspase-3 and sucrose water consumption in model group and PTX group have significant relationship (P<0.05 or P<0.01). And the bivariate related of IL-6 and Caspase-3, IL-6 and Caspase-8, TNF-α and Caspase-3, TNF-α and Caspase-8 have no relationship (P>0.05) in Sham group. And the bivariate related of IL-6 and Caspase-3, IL-6 and Caspase-8, TNF-α and Caspase-3, TNF-α and Caspase-8 in model group and PTX group have significant relationship (P<0.01).Conclusion:After myocardial infarction operation, the rats shown many depression symptoms.The rat show decrease activity, interests decreased in new environment, deficiency of pleasure, deficiency of exploration spirit, and behavioral despair increased, but the quantity of inflammatory cytokines and the protein levels of apoptosis were increased. The inflammatory cytokines in the brain issue HE staining increased and brain issue significantly disorder. After the treatment with PTX, the rats increase activity and decrease the depression behavior and the quantity of inflammatory cytokines and the protein levels of apoptosis. The inflammatory cytokines in the brain HE staining were decreased. All above the results indicate the inflammatory blocked could improve the depression symptom in rat after acute myocardial infarction. The production of depression after acute myocardial infarction because of the apoptosis in the limbic system of brain. This experiment indicate the inflammatory produced after rat acute myocardial infarction into blood- brain barrier, then initiating apoptosis in limbic system of brain, producing the depression after acute myocardial infarction behavior. According to the result of this experiment, inflammatory is the mechanism of depression after acute myocardial infarction. The PTX is the inhibitor of inflammatory factor, the rat depression symptoms improvement and the quantity of inflammatory cytokines TNF-α, IL-6 decreased in both serum and brain and the protein levels of apoptosis Caspase-3 and Caspase-8decreased and also the improvement of brain issue and the inflammatory cytokines in the brain issue HE staining further certificate the relationship between the depression after acute myocardial infarction and the inflammatory in other side. And bivariate related analysis, it is concluded that there is no relationship in inflammatory and sucrose water consumption, apoptosis and sucrose water consumption without acute myocardial infarction in Sham group.while the inflammatory and sucrose water consumption, apoptosis and sucrose water consumption have significantly relationship in model group and PTX group, which proved inflammation factors and apoptosis factors causing rat depression behavior after myocardial infarction. And bivariate related analysis, it is concluded that there is no relationship in inflammatory and apoptosis without acute myocardial infarction in Sham group, while the inflammatory factors and apoptosis have significantly relationship in model group and PTX group, which proved inflammation factors causing myocardial infarction after brain mechanism of apoptosis in the limbic system to make the body produce the causes of depression...
