Coreopsis tinctoria Nutt. is an annual herb of compositae coreopsis, Whose grass named Sanvitalia procumbens Lam. The dried inflorescences called snow chrysanthemum. In flora of Xinjiang, snow chrysanthemum was called Gulqai. People in Xinjiang also called it snow chrysanthemum, Kunlun snow chrysanthemum, Tianshan snow chrysanthemum, etc.. Coreopsis tinctoria Nutt. has the function of clearing heat-toxin, promoting blood circulation for removing blood stasis, harmonizing stomach and strengthening spleen. Snow chrysanthemum is the rare alpine wild plant with unique effect and high medicinal value with the snow lotuses in Xinjiang. Modern pharmacological research revealed that snow chrysanthemum has many flavonoids, essential oil and polyaeetylenes. It can obvious hypoglycemic, reduce blood pressure, reduce hematic fat, antioxidant and has other pharmacological activities. Our research group firstly found polyacetylenes from Chrysanthemum though systemicchemical composition studies. And finding that snow chrysanthemum has significant activity to treat inflammation, which is the important reason of coronary atherosclerotic plaques, by active experimental screening in vitro. Therefore, polyacetylene in snow chrysanthemum is likely the material basis to preventive and cure cardiovascular diseases. Howere, the main compounds of the polyaeetylenes are remainly unknown. It is inherent urgency to carry out this research. So, this paper research on the enrichment process and chemical characterization of the polyaeetylenes in snow chrysanthemum.1. The enrichment process of the polyaeetylenes was studied and optimized with mono-factorial experiments and orthogonal design.The optimal enrichment conditions were as follows:AB-8 macroporous resins were used for enriching the polyaeetylenes in snow chrysanthemum. The largest sample amount was 3.5BV of extracts of snow chrysanthemum (0.128g/ml). The eluting solvent was 90% methanol, the ratio of diameter to height was 1:10 and the percolate rate was 3BV/h.2. To separate and purify the polyaeetylenes compounds by column chromatography and semi-preparative HPLC. Finally, six polyaeetylenes have been obtained and the structures of them have been elucidated by H’-MNR and C13-NMR.They were (3S)-(6E, 12E)-tetradecadiene-8,10-diyne-1-ol-3-O-β-D-glycoside, coreoside A, (E)-7-phenyl-2-heptene-4,6-diyn-l-ol, (Z)-7-phenyl-2-heptene-4,6-diyn-l-ol, (2S)-(3Z,11E)-decadiene-5,7,9-triyne-1,2-diol,(2R)-(3E,5E, 11E)-triene-7,9-diyne-1,2-diol.3. This study employs the technology of HPLC to characterize and identify the polyaeetylenes. As a result, we established the HPLC fingerprint of the polyaeetylenes. Identification and analysis of the polyaeetylenes, and we successfully identified 3 main constituents in snow chrysanthemum. This study employs the technology of HPLC to characterize the content determination of polyaeetylenes in snow chrysanthemum. As a result, we decided the content determination of coreoside A, (3S)-(6E,12E)-tetradecadiene-8, 10-diyne-1-ol-3-O-β-D-glycoside, (E)-7-phenyl-2-heptene-4,6-diyn-l-ol.The content of them are 0.2439 mg/mg、0.070 mg/mg、0.0068 mg/mg, respectively.4. This study investigated the effects of polyaeetylenes on the proliferation of 3T3-L1 pre-adipocytes and tested two polyaeetylenes compounds for their in vitro anti-inflammatory effects. The anti-inflammatory action attributable to the inhibitory action of LPS-induced NO production in BV-2 cell.
