Study On The Difference Of Plasma MicroRNA Expression Between Knee Osteoarthritis And Normal Human

1 Objective:we base on Chinese medicine theory of preventive treatment of diseaseto comparing differential expression of miRNA in normal and osteoarthritic patients serum, and screening the miRNA that up-regulated in osteoarthritic serum. We will use the experimental result to diagnose the osteoarthritic earlily or judge the osteoarthritic prognosis.We will bring new discovery to preventive treatment of disease of Chinese medicine.2 Methods:Selected 13 normal volunteers and 13 wangjing hospitals osteoarthritic patients serum as the specimen.via the world’s most advanced RT-PCR method to analysis accurated the length distribution of miRNA, compare the differential expression of miRNA in each specimen, painting the specimen miRNA scatter plot,In order to obtain credible molecular sequence which we need, in the process experiment we remove the join、polluted and low-quality molecular sequenceunder.The we statistic the credible molecular、the condition that fold change>l and P-value<0.01, dressing by screening the miRNA that maybe the pathologic gene of osteoarthritis.3 Results:Contrast the experimental specimen, we find many miRNAs were high expression in the osteoarthritis patients.Those miRNAs which were high expression is this research purpose.According to statistics,the condition that fold change>1 and P-value<0.01, dressing by screening the miRNA that is we need.We find five miRNAs are highly expressed in the osteoarthritis patiens.The five miRNAs are hsa-miR-378、hsa-miR-1268a、 hsa-miR-483-5p、 hsa-miR-375 and hsa-miR-1275.4 ConclutionAccording to statistics,hsa-miR-378、hsa-miR-1268a、hsa-miR-483-5p、hsa-miR-375 and hsa-miR-1275 were significantly up-regulated in the osteoarthrtic serum.they were maybe the pathologic gene of osteoarthritis.Experimental result can provide new technology lo early diagnosis the osteoarthritis.The result bring new discovery to preventive treatment of disease of Chinese medicine.
