Osteoarthritis (OA) is a kind of common chronic, multiple bone and joint disease in clinic. Much-of this disease occurs in the middle aged and elderly people,and through a large number of survey showed that the number of patients with osteoarthritis with age showed a gradual increase trend. The main clinical manifestations of this disease are joint stiffness, pain, flexion and extension. The degeneration of articular cartilage and subchondral bone destruction and osteophyte formed as the main pathological features. Various methods of treatment of the disease, However, the etiology, pathogenesis and treatment mechanism is not completely clear.This paper is divided into two parts:literature review and experimental study. Literature review from two aspects of modern medicine and traditional medicine, the etiology and pathogenesis of osteoarthritis, treatment and other aspects were reviewed. At the same time, the relationship between MMPs and osteoarthritis were reviewed, the study shows that the occurrence of osteoarthritis is related to many factors, especially the relationship between MMPs and the disease. As part of the study is to observe the changes of KOA model in rabbit serum MMP-3 and MMP-13 concentration of needle knife therapy in, and to explore the mechanism of acupotomy in the treatment of knee osteoarthritis.ObjectiveA large number of clinical studies show that Acupotomy is one of the effe ctive treatment of osteoarthritis therapy. In this study, the effect of needle knife the rapy in knee osteoarthritis rabbit model of MMP-3 and MMP-13 long-term content change.Preliminary study on the relationship between MMPs and osteoarthritis, in order to explain mechanism of acupuncture therapy in the treatment of knee osteoarthritis, To provide experimental basis for clinical application of loosening therapy in the treatment of knee osteoarthritis with acupotomy.Methods48 healthy New Zealand rabbits(Six-month-old, male and female, weight 2.25+-0.1 kg) were randomly divided into four group,normal control group, model group, electro-acupuncture group and acupotomy group. After normal group not to participate in the building, The remaining three groups with the left hind leg unbend a fixed braking method experimental rabbit model of knee osteoarthritis. Model group not to intervene, The electro-acupuncture group intervention 3 times a week, The Acupotomy group Intervention treatment once a week, Both group intervention for three weeks. After 3 weeks of intervention, using enzyme-linked immunosorbent assay(ELISA) method to detect model rabbit serum concentrations of MMP-3 and MMP-13, analysis of effectof acupotomy therapy on the content of MMPs in serum.ResultsThe results of ELISA detection of MMP-3 and MMP-13 in rabbit KO A serum showed that the content of MMP-3 and MMP-13 in model group was significantly increased, Compared with other groups, the difference was statistically significant (P<0.05); MMP-3 and MMP-13 content in serum of acupotomy group decreased significantly and the difference compared with the model group and EA group was statistically significant (P<0.05).ConclusionAcupotomy therapy can reduce the concentration of MMP-3 and MMP-13 in knee osteoarthritis model in rabbit serum.Therefore, acupuncture therapy can reduce the degradation of the extracellular matrix of cartilage, and it can inhibit the destr uction of articular cartilage and control the degeneration of cartilage. To improve the symptoms of osteoarthritis of the knee, the role of the treatment of KOA.
