According to traditional knowledge represent method ,the knowledge resource lacks a uniform semantic description. It is hard for user to find more relevant knowledge resource and realize the information semantic fusion. How to assign the managed knowledge resource a machine understandable meaning to facilitate user to find what they really need ,and according to the domain knowledge to realize the information semantic fusion ,are challenging tasks for knowledge represent.The domain ontology is a formal, explicit specification of a shared conceptualization , which can represent domain knowledge and facilitate the interoperation between the heterogenous application systems. Ontology is a kind of conceptualized description towards domain knowledge which reflects experts' opinions on this field's knowledge. The ontology can not only support content enquiry of bottom database, but also represent the relevance of database through statemental description about semantic information which not dependent on specific date organization pattern.After analyzing the shortcomings of traditional knowledge represent, the article probes the ontological knowledge represent and investigate the description logic further, then analyzing the reason of ontology language on the basis of description logic.The ontology has many different represent patterns, that is , ontology description language. Different ontology usually has different logical structural organizations. In a more informational system environment, each isolated developed system has it's own internal present model and ontology. Therefore information exchange between these systems should be realized through a specific knowledge aggregation and procession mergence which also means the ontology mapping. Element mapping of ontology can be achieved through analyzing grammatical and semantic conceptual similarity. Then the result of ontology mapping will be distributed to every knowledge datebase and carry on reasoning and searching.At last, a knowledge reasoning and searching system prototype based on ontology is put forward .Meanwhile we also gives application of inquiry system in teaching and education field .Knowledge represent in this system expresses with the form of triple and consumers can inquire by selecting. Consumers' requirements collected by client will be transferred to server and the inference engine will hand them after getting the information from the server, then clients can get the corresponding knowledge.
