This paper reported an improved approach to prepare the silica sol-gel film. By the optimized condition of sol-gel process,a nano-scale thin-film was prepared. The structure properties and the thickness of the film were also characterized. The chemically modified electrodes were studied that used the nano-scale silica sol-gel film as the matrix for immobilizing the molecules such as the methylene blue, the thionine or the alizarin red S on the surface of gold electrode. The sol-gel film showed the excellent chemical and electrochemical properties as well as kept the quality electrochemical activity of those immobilized redox molecules. The modified molecules took place the typical adsorption controlled electrochemical reaction, and furthermore, the modified methylene blue gave the obviously electrochemical catalytic affection on oxidation of NADH or reduction of hemoglobin if they were immobilized in the film meanwhile. Alizarin red S sol-gel CME can be used directly to determine the total aluminum in natural and drinking water.
