1,Explore the vertical jump pattern of movement effect on bone mineral density in the tail-suspended rats.2,Explore the vertical jump pattern of movement effect on biomechanical indicators in the tail-suspended rats.3,Explore the vertical jump pattern of movement effect on femur Runx2mRNA expression in the tail-suspended rats.【Methods】1,Establish rat vertical jump pattern of movement, the 18 4-week-old SD rats were divided into three groups, and have 8 each group, then test indicators after 8 weeks exercise and 2 weeks tail-suspension.2,Measure longth,distal thickness and width,proximal thickness and width,middle thickness and width of left femur and tibia with the vernier caliper.3,Scan femur and vertebrae bone mineral density with Hologic Discobery A DEXA.4,Test each biomechanical index with three-point bending test and compression test.5,Detect rats left femur metaphysis Runx2mRNA expression with real-time fluorescence quantitative PCR of dye method. At the same time, detect sample GAPDHmRNA expression quantity as internal reference of relative quantitation of sample target genes.【Results】1,Comparison of three groups of rats weight:The group of jump plus tail suspension and the group of swimming plus tail suspension has significant difference compaired with the group of quiet plus tail suspension(P<0.05), The weight of rats in each group has no significant difference after two weeks of suspension (P>0.05).2,Compare the morphological index of femur and tibia comparison: Several index of the group of jump plus tail suspension has increasing trend compaired with the group of quiet plus tail suspension, but has no significant difference (P>0.05).3,Compare the BMD of femur:The group of jump plus tail suspension has very significant difference compaired with the group of quiet plus tail suspension(P<0.05); the group of swimming plus tail suspension has no statistically significant difference compaired with the group of quiet plus tail suspension(P>0.05).4,Compare the BMD of vertebra:The group of jump plus tail suspension has significant difference compaired with the group of quiet plus tail suspension(P<0.05); the group of swimming plus tail suspension has no significant difference compaired with the group of quiet plus tail suspension(P>0.05), But there is a downward trend slightly.5,compare index of femur bone biomechanical comparison:The group of jumping plus tail suspension has no significant difference compaired with the group of quiet plus tail suspension(P>0.05); the group of swimming plus tail suspension has no significant difference compaired with the group of quiet plus tail suspension(P>0.05), but there are some downward trend.6,Compare index of spinal bone biomechanical:The group of jumping plus tail suspension has no significant difference compaired with the group of quiet plus tail suspension(P>0.05); the group of swimming plus tail suspension has no significant difference compaired with the group of quiet plus tail suspension(P>0.05), but downward trend slightly.7,Compare the level of Runx2mRNA in femur cartilage tissue:The group of jumping plus tail suspension has significant difference compaired with the group of quiet plus tail suspension(P<0.05); the group of swimming plus tail suspension has no significant difference compaired with the group of quiet plus tail suspension(P>0.05), But there is a downward trend slightly.【Conclusion】1,swimming have no significant effect on tail-suspended rats femur and vertebra bone mineral density,biomechanical index and femur Runx2mRNA expression.2,The vertical exercise have significant effect on tail-suspended rats femur and vertebra bone mineral density,biomechanical index and femur Runx2mRNA expression.3,Compared with the swimming, the vertical jump have more significant effect on prevent ability of bone metabolism decline because tail-suspension.4,Bone mineral density and maximum load about biomechanical index have significant correlativity5,vertical jump exercise was a good way of prevent osteoporosis, and have a good practical value for research of different movement pattern effect on bone metabolism.
