This study is designed to explore the role and mechanism of uphill and downhill running exercise on bone metabolism in ovariectomized mice, and analyse the impacts of these two exercises on bone metabolism. Ovariectomized mice by surgery is used to simulate postmenopausal women's bone mineral status, and we can analyse the impacts of uphill and downhill running exercise on bone mineral density, metabolic markers and mRNA expression of osteoblast-related factor in ovariectomized mice, and explore the reason,in order to provide a reference for the prevention and treatment of osteoporosis in postmenopausal women indirectly.Aim1. Establish the uphill and downhill running exercise model,in order to provide a reference for exercise and bone health related research.2. By ovariectomized mice model to simulate postmenopausal osteoporosis, to study the impact of exercise on osteoporosis in postmenopausal women.3. Comparative study between the downhill and uphill running exercises on bone mineral density and bone metabolic markers and osteoblast-related factors (BMP-2, Runx2, Osx, OCN) gene expression. in ovariectomized mice.Method32 C57 BL/6 female mice of 8-weeks-old were randomly divided into 4 groups: SHAM group, OVX group, OVX+UP group, OVX and DOWN group.The were 8 mice in each group. The mice of SHAM group were sham treatment, the ones of other three groups were ovariectomized. After 8 weeks of training, all the mice were slaughtered and tested bone mineral density, bone metabolic markers, and osteoblast-related factor mRNA expression.Result1. Compared with OVX group, the bone mineral density in metaphyseal tibia and femur is significantly increased in both OVX+UP group and OVX+DOWN group,especially obvious in OVX+DOWN group.2. Among blood biochemical parameters, the blood phosphorus of mice in each group has no significant changes; serum calcium and ALP in both OVX+DOWN group and OVX+UP group were significantly higher than in the OVX group.3. Among osteoblast differentiation-related factors, OCN of the mRNA does not change significantly; the content of BMP-2, Runx2, Osx mRNA in OVX+ DOWN group is significantly higher than the one in OVX group; the content of Runx2, Osx mRNA in OVX+UP group is significantly higher than the one in OVX group, and the content of BMP-2 mRNA in OVX+DOWN group is significantly increased than the one in OVX+UP group.Conclusion1. The uphill and downhill running exercises have obvious influence on tibial and femoral position BMD in ovariectomized mice; and the effect of downhill running exercise is significantly better than uphill running exercise.2. The uphill and downhill running exercise can significantly affect the serum calcium, ALP, and downhill running is better than uphill running; but the impact on phosphorus in the two exercises is not obvious.3. The downhill running can significantly increase BMP-2 mRNA expression; the effect of the uphill and downhill running on the Runx2, Osx mRNA expression is obvious, and the downhill running is better than the uphill running; the effect of the two exercises on OCN is not obvious.4. Both he uphill and downhill running exercises are healthy for bone,and the down-hill running exercises is more better.
