| I ObjectiveFree Aging Theory considered that the reason of senility is that body's oxidation and the balance of antioxidation were damaged. Fitting exercise can improve body's oxidation and the balance of antioxidation, and also have anti-aging effect. While, there are differences between fitting exercise and other ways in anti-aging. This research through increasing load movement for elderly rats myocardial tissue antioxidant enzymes activity, mitochondria changing patterns of gene expression and antioxidant enzyme gene transcription factors and antioxidant enzymes from three aspects of movement of aerobic exercise, delay ageing for cardiac muscle ageing delaying reveal the mechanism of sport, but also provides a new basis for some related diseases of aging provide new reference.II materials and mothodsLaboratory animal models:18 months,30 female and healthy Sprague-Dawley(SD) were randomly divided into two groups, and 15 in each group, control group (c:n=10,c:a=3months). All of those SD were fed in separate cage according to the experimental animal feed norm. Those rats were accept treadmill running training, controlling sports intensity with Application of Blood Lactate.Formulating middle-aged rats'increase aerobic exercise scheme, weekly exercise 6 times for 20 weeks.Rat's exercise mothod:sports training after the adaptability of rats, increasing load, speed running motion for 15 meters per minute, time for 30 minutes after the first increases, according to the training time and then the intensity of the principles are increasing, training time increased range for 5 minutes, run speed increase for 1 meter, each load lasts for a week.6 times per week training at 4:30, started training on Sunday to rest.Exercise method:after adaptive training, those rats were accept treadmill running training,the speed at the beginning was 15 meters per minute, time for 30 minutes, according to the training time and then the intensity of the principles are increasing, training time increased range for 5 minutes, run speed increase for 1 meter, each load lasts for a week,6 times per week training at 4:30PM for 20 weeks, and rest on Sunday.Test plan of Lactic acid valves:Using the method of cutting tail to collect blood, with lactic acid power curve for the principle, reference to Feng meiyun's Lactic acid valves treadmill running training to study. Initial load for 15 meters per minute, lasts for 2-3 minutes, increasing amplitude is 3 meters per minute, intermittent 30 seconds. After those rats were being trained for 24 hours, took blood from their's orbital,centrifugal take on 4 degrees Celsius in 20℃and-to save, refrigerator. Taking the rat's cardiac muscle, put half of this into liquid nitrogen in store, and others put into paraformaldehyde to keep.Using the cardiac muscle part with liquid nitrogen to identify myocardial tissue antioxidant enzymes activity, mitochondria changing patterns of gene expression and antioxidant enzyme gene transcription factors and antioxidant enzymes.ⅢResultsChemical result:1,The group's of young rats'and elder rats'SOD and GSH—Px activity are higher than elder and quite rats'. There are great difference between young rats and quite elder rats (P<0.01), elder exercise rats'SOD and GSH—Px activity higher than quite eider rats 0.05 percent.2,The group's of young rats'and elder rats'CAT activity are higher than elder and quite rats'. There are great difference between young rats and quite elder rats (P<0.05), but, there are no difference between elder exercise rats'and quite eider rats in CAT activity.3,The group's of young rats'and elder rats'MDA content are lower than elder and quite rats'. There are difference between young rats and quite elder rats (P<0.1). Elder exercise rats'MDA content higher than quite eider rats 0.05 percent.The results of examination by the methods electron microscope:Myocardial mitochondria in quite and young rats has an arrayed order, myocardial mitochondria are in good integrity, no cavitation and edema; myocardial mitochondria in quite and elder rats are swelling in different degree. Parts of them become Focal cavitation, ridge become less, forming Electronic translucent zone. Because of the excessive swelling, some myocardial mitochondria limitans and mitochondria are broken; myocardial mitochondria in exercise and elder rats are seldom swelling, and none of theirs'limitans is broken.The results of microarray detecting:After 20 weeks training of increaing aerobic exercise,from the comparision between the elder aerobic exercise group and the elder quite group,we can see the changs about myocardium and related genes of antioxidant enzymes in the elder aerobic exercise group as follow:Ggt1 is three times lower than before; S100g is 2.9 times higher than before; Kcnmal is 2.9 times higher than before.Conclusion:1,Increasing aerobic exercise can obviously improve elder rats'Myocar-dial antioxidant enzymes, such as SOD,GSH-Px and CAT activity.It can also imcrease NO content of the rats'myocardium,and decrease MDA content of the rats'myocardium.2,Increasing aerobic exercise can improve the structure of the Rats' Cardiac Muscle Mitochondria and can delay the changes of mitochon-dria's structure caused by aging. 3,There are no obvious changs which caused by the Increasing aerobic exercise in the elder rats'myocardium and related genes of antioxidant enzymes.The antioxidant enzymes'activity may cused by antioxidant enzymes'synthetic substrate or changes of some ion in regulating gene. |
