Purpose:1. Association between KCNA7 Gene T418M Polymorphism and elite endurance athlete (EEA) status.2. Association between KCNA7 Gene T418M Polymorphism and the effects of endurance trainingSubjects and Methods:Subjects:①EEA(elite endurance athletes): They comprise 123 unrelated Chinese elite endurance athletes. They were specialists in long-distance running including 5000m, 10000m and marathon.②NS(new soldiers): They consist of 102 unrelated Chinese man new soldiers from a police army.③SCON(sedentary control): They comprise 206 unrelated Chinese sedentary undergraduates and have no trained experiences, as well as their parents. All subjects are Han population and plain-men in the North of China.Methods: PCR-RFLP was used to examine the genotype and allele frequencies of two subjects; Sequencing was used to analyze all three genotypes. The NS undertook a distance running program consisted of 5000 m running with the intensity of 95-105% ventilatory threshold (VT), 3 times/week for 18 weeks. VO2max,running economy (RE),body composition and Left ventricular structure and function were measured before and after the protocol, and were used to explore the association between KCNA7 gene T418M polymorphism and the effects of endurance training.χ2 test was used to compare the frequencies between different group subjects, and to determine whether the genotype distribution differed from that expected under the Hardy-Weinberg equilibrium. ANOVA tested differences in variables before the training between the three genotypes, and Post-Hoc tested differences between two genotypes. One way ANOVA was used to analyze differences in change of variables between genotypes. P<0.05 were considered statistically significant. All statistical analyses were performed with SPSS software for Windows 11.5 package. The results are presented as means±SD.Results:⑴KCNA7 gene T418M polymorphism was C/T, and there were no significant differences between Chinese Han population and those in the population of northeast China.⑵Findings in the present study demonstrated that there were significant differences in genotype frequencies between the EEA and SCON, but no significant differences in allele.⑶Findings in the present study demonstrated that there were significant difference in genotype frequencies between male EEA and male SCON, as well as between the marathon athletes and SCON.⑷There were significant differences between gene polymorphism and BMI before training, but their changes after training were no significantly different. On the contrary, no associations were detected between gene polymorphism and LBW before training, but there were significant difference in the changes after training.⑸KCNA7 gene T418M Polymorphism might contribute to individual difference in the left ventricular function at 100w load.⑹No association was detected between KCNA7 gene T418M Polymorphism and VO2max before training, as well as their changes after training. There were significant differences in△VO2RE and△VO2RE/W after training between three genotypes, and the degree of the responses in VO2RE was related to the genotypes(p<0.05), with the largest in CT, then the TT and CC.Conclusion: The present study demonstrated that KCNA7 gene T418M polymorphism is associated with the elite endurance athlete status, and it might contribute to individual differences in RE and LBW to endurance training.
