Flavonoids are abundance in nature, especially in plants. It is estimated that about 20% herbs contain flavonoids. Many research have indicated that flavonoids had multi-biological-activity. For example, antibacteria, diminishing inflammation, anti-mutation, lowing blood pressure, antioxidation, curing and preventing cancer. Flavonoids can eliminate most of oxygen radical, they can improve the activity of SOD and reduce the amount of MDA. Due to multi-biology activity of flavonoids, more and more people begin to study flavonoids. Now flavonoids are not only used to prevent or cure much diseases, such as hypertension, hyperlipidmia, coronary heart disease, angina, but also broadly used in some industrial fields, such as cosmetic and foodstuff.It is considered that the fruit of Forsythia suspensa was a traditional Chinese herbal medicine, but the folk investigation showed that the leaves of Forsythia suspensa had the effects of eliminating constipation, preventing common cold, reducing blood lipid and so forth. Some studies showed the content of effective compounds in Forsythia suspensa leaves is higher than that in fruits. Forsythia suspensa were made into tea as nutritious drinking. Some studies in our lab showed that Forsythia suspensa leaves have good effects on antioxidation in vitro and in vivo, protecting liver, lowing blood pressure, and having fatigue resistance effect.The object of this study is to investigate the effect of Forsythia- leaf-flavonoids on fatigue resistance in swwiming-induced injury model in mice. In this experiment, we gained Forsythia-leaf-flavonoids by ethanol circumfluence and Polyamide column chromatography ,then we tested the effect of the extraction. 75 healthy ICR male mice were randomly divided into five groups, which are quiet group(QG) , swimming contrast group(SCG) , min-dosage FLF group(MinG), middle-dosage FLF group(MidG) and max-dosage FLF group(MaxG). The Mice of MinG, MidG and MaxG were injected through mouth with different dosage but the same volume Forsythia-leaf-flavonoids , QG and SCG were injected through mouth with 0.9% NaCl in the same volume for 30 days;Mice rest for 24 hours after them swimming to tired, then 40 of them were executed to adopt blood from eye socke, and to take out liver, heart, brain and musculus quadriceps femoris, then measured the content of haemoglobin, blood sugar, serum urea nitrogen, blood lactic acid, liver glycogen, muscle glycogen , and MDA content in liver, heart, brain and musculus quadriceps femoris,tested the activity of catalase in blood ,SOD activty in liver, heart, brain and musculus quadriceps femoris, and Cu/Zn-SOD activity in liver and heart. We took out the liver and heart from the other 35 mice, and performed immunohistochemical detection of Cu/Zn-SOD, and observed the ultrastructure of the same position of liver, heart, musculus quadriceps femoris of different mice by transmission electron microscope.The results of this experiment show that:1. Forsythia- leaf-flavonoids (FLF) yield is 8.48 % ,the flavonoids content in it is 49.2%.2.FLF can obviously prolong the time of swimming, especially MaxG and MidQtheir rations are 32.4% and 29.8% respectively.3.FLF can enhance mice liver glycogen and muscle glycogen content, reduce serum urea nitrogen and blood lactic acid content after mice swimed and rest for 24 hours.4.FLF can enhance the activity of serum cataiase and'^SQD, reduce the content of MDA in liver, heart, brain and musculus quadriceps femoris.5.FLF can enhance the activity of Cu/Zn-SOD in liver and heart, increase the expression of Cu/Zn-SOD gene in liver and heart.6.The observing of ultrastructure by transmission electron microscope showed that FLF had the effect of protection in liver, heart and musculus quadriceps femoris in swwiming-induced injury model. In SCG, cell was injuried in liver, heart and musculus quadriceps femoris as a result of fatigue swimming, in MaxG, the result was close to QGConclusion: The result indicated that it is abundant of flavonoids in Forsythia Suspensa leaves. FLF can enhance mice sport ability and promot the restoration of fatigue mice. Perhaps the effects of FLF are related with its anti-oxoidation activity.
