This study was designed to establish the sports anemia rats model and evaluate the effects of training on sports anemia and nutrition supplement on rat brain iron metabolism.Experiment design: including two parts: Sports anemia model establishment and nutrition supplement support. Partâ… : Thirty Wister males rats (4 weeks old) were randomly divided into two groups: control group (C, n=10) and exercise group (E, n=20). Group E rats were trained by running on treadmill for 5 weeks in progressive increasing workload. At first, the grade was set to zero degree and the speed was set to 30 m/min. The training was taken from Monday to Saturday, no training on Sunday. The rats were trained once a day in the first two weeks and twice a day in the last three weeks in the morning and evening respectively. Training Plan: The first training duration was 1 minute, then since the second time, the duration was two minutes longer than that of the last one. The grade and speed kept constant. When 5 weeks' training had been finished, blood was taken from the tail of all rats to analyze the level of HB, RBC and HCT. Part â…¡: From the sixth week, the rats in Group E were divided into two groups: exercise group (EE, n=10) and exercise with nutrition supplement group (EN, n=10). Group EE and EN rats went on running on treadmill for another 6 weeks, with the same grade and speed as Part â… . The training was taken twice a day from Monday to Saturday, no training on Sunday. The training duration of the sixth week was 80 minutes and 85 minutes in the seventh week. From the eighth to the eleventh week, the duration increased by 2 minutes a day since 90 minutes. After each training, EN rats were given nutrition of the anti-sports anemia complex. When 11 weeks' training completed, all rats were killed for the following measurements: HB, RBC, HCT, brain iron, brain ferritin and brain transferrin concentration.The results of the experiment showed: After 5 weeks' treadmill running training, the HB, RBC and HCT level of Group E rats were significantly lower than those of Group C (p<0.01). After 11 weeks' training, the HB and RBC level of Group EE were significantly lower than those of Group C (p<0.05) and Group EN (p<0.01, p<0.05). Both brain iron and transferrin concentration did not show significant differences between groups. The brain ferritin level of Group EE was significantly lower than that of Group C (p<0.05), whereas no difference compared with Group EN. There was also no difference between Group EN and Group C in rat brain ferritin concentration.The results suggested that five weeks' treadmill training could cause sports anemia in rats and six weeks' nutrition supplement could improve the situation of sports anemia obviously. The brain ferritin level of rats with sports anemia decreased significantly, which suggested that sports anemia could result in brain iron metabolism variations in some ways. Six weeks' nutrition supplement could improve brain iron status of sports anemia rats.
