| Object To observe the change of Cycloxygenase-2 (COX-2),hypoxia inducible factor-1α(HIF-1α),bcl-2 staining in myocardial cell round the wound area heart of rat after experimental severe myocardial contusion and the relationship of the change and injury time with immunohistocheimistry,and to clarify the molecular mechanism of severe myocardial contusion.Method A experimental severe myocardial contusion model was established, and using SABC immunohistochemical and computer image analysis, expressions of COX-2,bcl-2,HIF-1α protein after severe myocardial contusion in different time were evaluated. Results the control-experiment is negotive, the expressions of COX-2, bcl-2 and HIF-1α could be observed at 6h after severe myocardial contusion, and then increased with the prolonging of time , then got to a peak at 12h,12h and 2d after injury respectively, by 6d the positive staining cells still could be detected. Conclusion The model of severe myocardial contusion in rats has been set up successful; COX-2, bcl-2 and HIF-1α may play a role in severe myocardial contusion; as an objective indication, the time sequence regularity of COX-2, bcl-2 and HIF-1αafter severe myocardial contusion may be applied in severe myocardial contusion time estimation. |
