With increasing of acknowledgement about the moleculopathology of central nervous system after injury and the application of immuno-histochemical techniques, a lot of studies have done on the relationship between the cerebral contusion and its aging in respent of immu-nohistochemistry. Limited by the human material, the studies are mainly imitative experiments of animals 'cerebral contusions, which have committed the diversity between the theological study and the practical identification.At present, by the immunohistochemical techniques, there have been the expressions of HSP70, bFGF, COX2 after rat cerebral contusion. The consequences show that the expressions of these three indications show certain changing regulations after rat brain contusion. But there has not been any report about the application of these indications on the estimation of the aging on human cerebral contusion. So this study will reveal the human expressions of HSP70, bFGF, COX2 after cerebral contusions and the relationship between the expression and the contusion time utilizing the immunohistochemical staining, At the same time, this study will find out the diversity between the autopsy case and the animal experiment, which can provide the theorical evidence to identify the aging cerebral contusion.Materials and methods1. Experimental Group: choose 35 autopsy cases during 1995 -2001, in which deaths are caused by closed cerebral injuries with obvious cortical contusion foci. Divide them into seven groups. Choose 5 cases as control, which died from immediate deaths, and not caused by cerebral injuries. The age of cases vary between 19 to 58, with average age of 38.09 and without sex choice.2. The methods of immunohistochemical staining: After removing paraffin in xylene and rehydrating through graded alcohol, sections were treated with 3% hydrogen peroxide to inactivate endogenous. This was followed by antigen retrieval by microwave heating. Sections were blocked by normal goat serum for 20 minutes at room temperature and incubated for 1. 5 hours at 37℃ with polyclonal antibodies to, re-spectively, HSP(1:100), bFGF(l:100), and COX2(1:50). After incubation with the appropriate secondary antibody and streptavidin -conjugated peroxidase complex or streptavidin biotin complex, the im-munocomplex was visualized by using diaminobenzidine as a chromo-gen in a peroxidase reaction. Between each steps washed in PBS three times for 5 minutes, with the exception of removing the normal serum. After dehydrating through graded alcohol and xylene, sections were mounted on gelatin coated slides and coverslipped.3. The gray value of positive nerve cells were detected by image analysis system. Then the data analyses were performed using SPSS for Windows 10. 0 with a significant level of P <0. 01.Result1. In immediate death group, the gray gradient of HSP70 in nerve cells was the smallest, which had obvious difference with other experimental groups (P <0. 01). As the contusion time prolonging, the gray value of HSP70 gradually increased, and reached the peak pointin the group of 20 - 27 h, which had magnificant difference with other groups except the group of 10. 5 -11.5 d. Then in every experimental group, the gray values of HSP70 slightly decreased, while it increased again in the group of 10. 5 - 11. 5 d.2. In immediate death group, the gray value of bFGF positive nerve cells is the smallest, which had magnificant difference with other experimental groups ( P < 0. 01 ) . The gray value of bFGF in the group of 10 - 14 h abruptly increased ( P <0. 01). Then the gray value of bFGF increased slowly and reached the peak point in the group of 10.5-11.5 d.3. In the group of 10 - 14 h, the gray value of COX2 positive nerve cells began to decrease obviously (P <0. 01). Then reached the lowest point in the group of 20-27 h, which had magnificant difference with other groups ( P < 0. 01).
