Research On The Effective Fractions Of Kudiezi Injection

Kudiezi Injection is made from abstractions of Ixeris sonchifolia(Bge.)Hance, mainly used in treatments for coronary heart disease and angina pectoris, carrying the functions of distending the blood-vessel, improving Blood oxygen saturation supply in Myocardium, increasing Fiber protein haemolytic enzymes activity, inhibiting thrombosis, etc. No untoward effects have been found through years of clinical application,and curative effects have been proved.Chemical compositions of Kudiezi Injection have been researched in the project. Six compounds have been distracted from the preparation, two of which confirmed to be organic acids, while the other four fixed to be Flavones by testing results of TLC, 1H-NMR and 13C-NMR. The organic acids are (-)-(2R,3R)-3,4-di-hydroxyl-phenylacetyl tartrate(CompoundⅠ) and chicoric acid(CompoundⅡ). The Flavones are luteolin-7-O-β-D-glucopyranosyl- (1→2)-β-D-glucopyran osid(CompoundⅢ), luteolin-7-O-β-D-glucopyranoside(CompoundⅣ), luteolin-7-O-β-D- glucuro-nopyranoside (CompoundⅤ) and Apigenin-7-O-β-D- gluco- pyranoside(CompoundⅥ). The testing result proves that Kudiezi Injection contains Organic acid and Flavone. The Injection has been separated into 40% ethanol concrete and 60% ethanol concrete by macroporous resin. The content of organic acids in 40% ethanol concrete is measured to be 92.28%, while the content of flavones in 60% ethanol concrete is 98.17%, which proves that the former is the organic acid part of Kudiezi Injection and the latter is the flavone part of it. The pharmacological experiment shows that Organic acid and Flavone could offer an obvious improvement on the acute myocardial ischemia electrocardiogram changes of rats triggered by Posterior Pituitary intravenous injection, and effectively give protection during the process, which preliminarily proves that Flavone and organic acid are the active principle in Kudiezi Injection.Two types of compounds are isolated from Kudiezi Injection, purified and researched on pharmacodynamics. Active principles have been found, adding technology content to the product, which builds up a solid foundation for the effectiveness, safety and re-evaluation of Chinese Traditional Medicine Injection.
