ObjectiveTo study the expression of nm23H1 and S100A4 in colorectal carcinomas tissue and their relationship with Pathological clinical features. To explore their correlation and relationship with invasion and metastasis in Colorectal cancer.Material and MethodThe paraffin specimens were taken from 58 patients with radical surgical resection of colorectal cancer in the Second Affiliated Hospital of Nanchang University from Jan.2008 to Jun.2009. A total of 58 cases as the experimental group, to record the detail clinical pathology, 20 normal intestine specimens were used as the control group. EliVisionTM plus immunohistochemical method was used to detect the expression of nm23H1 and S100A4 in colorectal carcinomas tissue and normal intestine tissue.Results(1) The expression of nm23H1nm23H1 was chiefly located in cytoplasm. The protein expression of nm23H1 was more higher in normal intestine tissue than colorectal carcinomas tissue, the obvious difference was significant. The expression of nm23H1 was not correlated with the age of patients, the gender of patients, the tumour size, liver metastasis or the location of tumor (P >0.05);The expression of nm23H1 were significantly correlated with infiltration degrees and lymph node metastasis (P<0.05).but the expressions of nm23H1 was significantly increased in TNMⅠ/Ⅱcompared to that of TNMⅢ/Ⅳ(P <0.05).(2)The expression of S100A4S100A4 located in cell nucleus. There was obvious difference of S100A4 positive rate among colorectal carcinomas and normal intestine tissue, and they also have a statistical meanings .The expression of S100A4 was not correlated with the age of patients, the gender of patients, the location of tumour, the tumour size , liver metastasis and TNM grade(P >0.05). The expression of S100A4 were significantly correlated with infiltration degrees and lymph node metastasis (P<0.05). The expressions of S100A4 and the differentiated grades of tumor tissue were negatively correlated, weaker differentiation tissue had higher S100A4-positive rate (P<0.05).(3)The relationship between the expression of nm23H1 and S100A4 in the colorectal carcinomas .When the expression of nm23H1 was negative ,S100A4 protein negative, moderately positive and strong positive for five cases, fifteen cases and fourteen cases in the colorectal carcinomas, respectively. When the expression of nm23H1 was moderately positive, S100A4 protein negative, moderately positive and strong positive for twelve cases, four cases and two cases. When the expression of nm23H1 was strong positive,S100A4 protein negative, moderately positive and strong positive for three cases, one cases and two cases. using semi-quantitative method to analyse the protein expression of nm23H1 and S100A4 in colorectal cancer in 58 cases. And using Spearman rank correlation analysis, There was a negative correlation between the protein expression of nm23H1 and that of S100A4 (r_s′=-0.402,P =0.002).Conclusion1. The expressions of S100A4 and nm23H1 correlated well with infiltration degree and lymph node metastasis in patients with colorectal carcinomas.2. The expressions of nm23H1 in patients with colorectal carcinomas was significantly increased in TNMⅠ/Ⅱcompared to that of TNMⅢ/Ⅳ.3. The expressions of S100A4 and the differentiated grades of tumor tissue were negatively correlated, weaker differentiation tissue had higher S100A4-positive rate4. There was a negative correlation between the protein expression of nm23H1 and that of S100A4.Specific mechanism is not clear, needs to be further study.
