Objective:Stem cells play a very important role in liver injury and liver regeneration. The "more derived" and "plasticity" of stem cell are the theories of transplantation for treatment the acute and chronic liver damage. The SDF-1/CXCR4 biological axis mediates bone marrow stem cells homing to the multiple tissues and organs during the regenerative repair after injury. The combination of SDF-1 and stem cell therapy plays an important role for liver injury in a new ways. In this experiment, we construct acute liver injury models of SD rats with carbon tetrachloride. We observe the repair of liver injury through liver function, survival and liver histological changes after the transplantation of bone marrow mononuclear cells (BMMNC).And then we use the incubated BMMNC with the specific receptor antagonist AMD3100 of CXCR4 to study the effect of bone marrow mononuclear cells homing to the injuried liver and repair it through the biological axis SDF-1/CXCR4. It provides a new target for therapy.Methods:The SD rat model of liver injury induced by carbon tetrachloride is established. The degree of liver injury is evaluated according to the mortality of rats, pathology morphology and the levels of biochemistry to find the best project. Based on it, the BMMNC are segregated by donor rats. The rats of liver injury are randomly divided into BMMNC transplant group and AMD3100 intervention group, the liver injury control group. The BMMNC are transplanted through tail vein. One week after transplantation, to test the liver function by taking blood and observe the mortality of rats, while taking the rat liver, hematoxylin-eosin (HE) staining of pathology morphology.Results:(1) Different extent liver injury can be induced by different dosages of carbon tetrachloride, and the pathophysiology of acute liver injury can be copied very well at the dose of 0.45ml/100g, and the model is very steady. (2)One week after transplantation, the mortality of the BMMNC transplanted rats is about 17%, while the mortality of the AMD3100 intervention group is about 83%.It is significant difference between the two groups(P< 0.05).The liver function of the BMMNC transplanted rats is lower than the liver injury group and the AMD3100 intervention group.Conclusion:(1)Acute liver injury of rats can be induced with carbon tetrachloride at the dose of 0.45ml/100g very well. (2)BMMNC play an important role on the treatment of the liver injury, AMD3100 inhibits the BMMNC homing to the liver injury.
