Objective: To study the engraftment, growth and differentiation of porcine autologous bone marrow mononuclear cell after being intracoronary transplanted into AMI region at different times (2 hours after AMI and 1 week after AMI).Methods: Bone marrow were aspirated from the ileum and bone marrow mononuclear cells(BM-MNCs) was isolated by Percoll gradient centrifugation .And BM-MNC was prelabeled with red fluorescence cell linker PKH26-GL. A 90-minute left anterior descending artery ligation followed by reperfusion was used to produce myocardial infarction. 8-10×10~7BM-MNCs or culture medium of the same volume were intracoronary infused into infarction related artery at 2 hours or 1 week after AMI. Specimens were harvested 6 weeks after AMI for histological study including HE, PTH, immunochemical stain of factor VIII and Desmin, and transmission electromicroscope study. Result: In the specimens of two intracoronary infusion groups, labeled cells were found only in the infracted area and not in the normal tissue.
