Effect To The Expressions Of Survivin And MMP-2 In Carcinoma Tissue By Intraoperative Regional Arterial Infusion Chemotherapy On Gastric Cancer

Objective: Gastric carcinoma is one of the most common malignant tumors in our country. Its incidence is about 50% in digestive tract tumors, and its mortality ranks first in malignant tumors. It seriously harms to human health. So in this experiment, intraoperative regional arterial infusion chemotherapy (RAIC) is to be carried out, and it is to pseudo-evaluate its effect and mechanism on gastric cancer cells.Methods: It was configured 100ml with normal sodium including 5-fluorouracil (5-Fu 1000mg/m~2) and cisplatin (DDP 40mg/m~2); which was slowly perfused by main supply artery of gastric tumor during radical resection of gastric cancer. The expressions of Survivin and MMP-2, in 36 cases of advanced gastric cancer, before and after the treatment of intraoperative RAIC, were shown by SP immunohistochemical staining, and pathological changes of tumor were detected.Results: Survivin and MMP-2 were expressed in the cytoplasm of tumor cells. In 36 patients, 23 cases showed positive expression of Survivin, 27 cases showed positive expression of MMP-2, and their positive expression rates respectively were 63.9%, 75.0%. After the intraoperative RAIC, the average optical density (AOD) of Survivin and MMP-2 were significantly lower than those before treatment, and their differences were statistical significance (P<0.05) in tumor tissue. After intraoperative RAIC, light microscope examination revealed mild hydropsia of intercellular substance, slight changes of karyopyknosis and coagulation of cytoplasm in cancer cells, invasion of inflammatory cells, and mild vasculitis in some of that cases.Conclusion: The intraoperative RAIC for gastric cancer could obviously depress the expressions of Survivin and MMP-2 and cause the histopathological changes of cancer cells. Accordingly, it has active significance to inhibit invasion activity of cancer cells, and prevent intraoperative iatrogenic spread of cancer cells and postoperative recurrence and metachoresis.
