The apoptosis rate of HeLa cells was analyzed by FCM using PI staining. The expressions of caspase-3 and Mcl-1 proteins were analyzed by western blot. Results: Typical morphologic changes of apoptosis in HeLa cells could be observed after treatment with AFMC and ChR at various concentrations of 0.1, 1.0, and10.0μg/mL by fluorescence microscope using AO/EB fluorescence staining. Analysis by FCM showed that the apoptosis of HeLa cells was induced by AFMC in a concentration-dependent manner and apoptotic effect of AFMC was stronger than ChR at the same concentration. Western Blot analysis showed that AFMC downregulated expression of Mcl-1 protein and upregulated expression of caspase-3 protein. Conclusion:1) AFMC could effectively induce apoptosis of HeLa cells cultured in vitro, in a concentration-dependent manner.2) Induction of apoptosis in HeLa cells was associated with up-regulating Caspase-3 protein expression and down-regulating Mcl-1 protein expression. | | Keywords/Search Tags: | Cervical cancer, 5-Allyl-7-Gem-Difluoromethoxychrysin, Bcl-2, Caspase-3, Apoptosis | PDF Full Text Request | Related items |
