Objective: The aim of this study was to investigate the effects of 5-Allyl-7-Gem-Difluoromethoxychrysin(AFMC) on apoptosis of human cervical cancer HeLa cells cultured in vitro,for providing a laboratory and theoretic evidence on searching for a new candidate for human cervical cancer chemotherapy.Methods: Cell apoptosis morphologic changes of HeLa cells were observed by AO/EB double fluorescence stain. The apoptosis rate of HeLa cells was analyzed by FCM using PI staining. The expressions of caspase-3 and Mcl-1 proteins were analyzed by western blot.Results: Typical morphologic changes of apoptosis in HeLa cells could be observed after treatment with AFMC and ChR at various concentrations of 0.1, 1.0, and10.0μg/mL by fluorescence microscope using AO/EB fluorescence staining. Analysis by FCM showed that the apoptosis of HeLa cells was induced by AFMC in a concentration-dependent manner and apoptotic effect of AFMC was stronger than ChR at the same concentration. Western Blot analysis showed that AFMC downregulated expression of Mcl-1 protein and upregulated expression of caspase-3 protein. Conclusion:1) AFMC could effectively induce apoptosis of HeLa cells cultured in vitro, in a concentration-dependent manner.2) Induction of apoptosis in HeLa cells was associated with up-regulating Caspase-3 protein expression and down-regulating Mcl-1 protein expression.
