Obiective Study the effect of LMWH administration on the pancreatic microcirculation of ANP rats with different time point, observe the survival time of rats, analysis the survival rate of ANP rat, research the influence of LMWH on SAP evolution. And thus provide a theoretical basis and experimental foundation for clinical improvement of the pancreatic microcirculation of SAP rats.Methods 160 healthy SD rats (weighed 250-300g) were divided into there groups randomly: LMWH group, n=64, NS group, n=64, S group: n=32;according to different time (0h,6h,12h,18h),L and N group were each divided into four subgroup randomly, L0h group (n=16),L6h group (n=16),L12h group (n=16),L18 h group (n=16),N0h group (n=16),N6h group (n=16),N12h group (n=16),N18h group (n=16)。ANP models were set up by injection of 5% sodium taurocholate (0.1ml/100g)under pancreatic capsule with A and B groups. The rats of SO group were performed operation to touch the pancreas only. The LMWH were administrating on ANP models in different time, L0h group (0h),L6h group (6h),L12h group (12h), L18h group (18h),and repeated each six hours,L18h(18h) group inject only once. And the control groups were administrating N only. 24h later, we observe the ascites, saponified spots, punctiform or lamellar hemorrhage and necrosis on pancreas, and the concentration of amylase, calcium, ET-1, NO, TXA2, and PGI2 in each group were detected, HE stain and electron microscopic observation were made, the pancreas histopathology score were calculated. The rest half of the ANP rats were keep feeding. Observe the survival time of rats, analysis the rate of rat survival in 72h. Results 1)Celiac general situation:24h after ANP rats were made,half of each group were laparotomized: yellow and slightly opacity hemorrhagic ascites, saponified spots, punctiform or lamellar hemorrhage and necrosis on pancreas could be found in L group. Light hemorrhagic ascites, saponified spots, lamellar hemorrhage and necrosis of pancreas could be observed in N group. There no such changes in S groups.2) Experimental index detection: Ca2+ and AMY: The concentration of calcium in L groups were accordingly higher than that in N groups, (P<0.01), the concentration of amylase in L groups were accordingly higher than that in N groups, (P<0.01), There is no statistic significance within each group.PAF: The concentration of PAF in L groups were accordingly lower than that in N groups, (P<0.01),and L0h(6.559±0.748),L6h(6.369±0.712)w e re lower than L12h(7.323±0.323)(P<0.05),L18h(7.343±0.615)(P<0.05); there has no statistic significance between L0h(6.559±0.748)and L6h(6.369±0.712), also no statistic significance between L12h(7.323±0.323)and L18h(7.343±0.615)(P>0.05).TXB2(TXA2),6-Keto-PGF1α:The concentration of TXB2(TXA2),6-Keto-PGF1 in L groups were accordingly lower than that in N groups, (P<0.01), TXB2(TXA2)(39.386±2.089)and 6-Keto-PGF1α(52.550±1.047)in L0h group and TXB2(TXA2)(40.220±2.338)and 6-Keto-PGF1α(52.374±1.604)in L6h were lower than TXB2(TXA2)(57.384±3.838)and 6-Keto-PGF1α(57.743±1.504) in L12h and TXB2 ( TXA2 )( 57.924±4.262 ) and 6-Keto-PGF1α( 58.093±1.189 ) in L18h(P<0.01); there has no statistic significance between L0hand L6h, also no statistic significance between L12hand L18h (P>0.05).ET-1,NO: The concentration of ET-1,NO in L groups were accordingly lower than that in N groups, (P<0.05),and L0h(81.753±7.227),L6h(80.358±8.270)were lower than L12h(92.971±6.849)(P<0.05),L18h(95.553±6.903)(P<0.01); there has no statistic significance between L0hand L6h, also no statistic significance between L12hand L18h (P>0.05). There is no statistic significance within each group.3 histological examinations:HE stain : N group: pancreatic acinar cells rupture, acinar structure disappears, lamellar hemorrhage and necrosis of pancreas could be observed, large number of neutrophil infiltration; L group: pancreatic acinar cells swelling severely, punctiform or lamellar hemorrhage and necrosis on pancreas, less normal pancreatic acinar cells and lots of neutrophil infiltration were observed. S group: pancreatic acinar structure exists , pancreatic acinar cells integrated, no hemorrhage and necrosis on pancreas, no neutrophil infiltration .The pancreas histopathology score: The pancreas histopathology score of group L {L0h(6.000±0.926),L6h(6.500±1.069),L12h(8.750±1.283),L18h(8.250±1.670)} was significantly Lower than group N{ N0h (10.000±0.756),N6h(10.250±1.282),N12h(10.500±1.195),N18h(8.250±1.670)} accordingly(.P <0.01),and the score of L0 group(6.000±0.926)and L6(6.500±1.069) group was lower than L12 (8.750±1.283)and L18(8.250±1.670) group(P <0.01), There is no statistic difference between L0 group ( 6.000±0.926 ) ,L6 ( 6.500±1.069 ) and L12 (8.750±1.283)and L18(8.250±1.670)group(P>0.05).Transmission electronic microscopy observation: N group: pancreatic acinar cell structure damaged, acinar structure disappears, lamellar necrosis tissues. Large amount of micro thrombus formation, blood vessel rupture, neutrophil exists in residual microvessel. L group: a few micro thrombus formation in blood vessel, a few pancreatic acinar cell exists . S group: there is no micro thrombus formation in blood vessel. pancreatic acinar cells and acinar structure intact.The pancreas histopathology score and the level of PAF,ET-1/NO,TXB2/6-Keto-PGF1α(TXA2/PGI2)in L group,N group,S group was positively linear correlated to the pancreas histopathology score respectively(r=0.347,0.435,0.518,P<0.01).The survival time of rats: the survival rates of group L48h and L72h were 71.87%and 46.87%,lower than SO group, (48 h: 100%,72 h :100%)(P<0.05)。Conclusion 1)In regulating the level of TXA2,TXA2/PGI2,ET-1/NO ET-1,NO in serum, and reducing the pathological injury to ANP rats, the administration of 0h(A1) and 6h(B1) is comparably more effective than 12h(C1) and 18h(D1).2)In regulating the level of ET-1,NO,TXA2,TXA2/PGI2,ET-1/NO in serum, and reducing the pathological damage to ANP rats, the administration of 0h(A1) and 6h(B1) is comparably more effective than 12h(C1) and 18h(D1).3)LMWH administration can effectively improve the pancreatic microcirculation of SAP rats and increase survival rate.
