Objectives:To study the effect of treatment with Xue Bi jing on the pancreatic injury in rats with severe acute pancreatitis, we could observe the levels of serum NO, MDA, SOD, IL-6, LPS and the pathology of pancreas, and it could provide theoretical ground for clinical.Methods:Fifty-four SD rats were randomly divided into three groups:Control group(group A), severe acute pancreatitis group(group B) and treatment group with Xue Bi jing(group C). Each group was divided into 12h and 24h subgroups (n=9 in each subgroup). SAP was induced by retrograde injection of 3.5% sodium taurocholate into biliopancreatic duct; Xue Bi jing (4ml/100g) was injected in abdominal after modeling success and which was given every 6 hours in the group C; Rats of group A were only operated with a celiotomy; Normal saline (4ml/100g) was injected in abdominal in the group A and B. Nine rats from each subgroup were killed. then the correlated indexes were evaluated at 12h and 24h after the model had been developed; The wet-to-dry (W/D) weight ratio of pancreas were measured; Part of pancreatic tissue were changed by HE staining; then the morphologic changes were evaluated; The weight of ascitic fluid in abdominal cavity was measured. The level of serum amylase, NO, IL-6, LPS, MDA and SOD in serum were measured. The data were analyzed with SPSS 11.5.Results: Observation of pancreatic tissue:â‘ Normal pancreatic tissue was observed in group A; Severe edema and local or lamellar necrosis of the pancreatic tissue were observed in group B; Mide edema and spotty necrosis were observed in group C.â‘¡M orphologic change: The clear structure of pancreatic lobules were observed, and there were no interstitial inflammatory cells and red blood cell in group A; Pancreatic lobules were severe destroyed, and alveolar structure was disappeared, Acinar cell atrophy, disappearance of cell nucleus, a large number of inflammatory cells and red blood cell were observed in group B; The structure of pancreatic lobules were broaden, a few inflammatory cells and red blood cell were observed in group C.â‘¢W/D weight ratio of pancreas in group B were found significantly higher than those in group A at two time points (P<0.05); In group C, W/D weight ratio of pancreas were significantly lower compared with those in group B at 12h and 24h (P<0.05).â‘£In group B, pathological injury was gradually aggratated with time was prolonged; The scores of pathological changes of pancreas in group B were significantly higher than those of group A at two time points (P<0.05). There was a significant difference between group C and group B at two time points (P<0.05).2.â‘ There was no ascitic fluid in abdominal cavity in group A; A large of bloody, the turbidity ascitic fluid in abdominal cavity in group B; A small amount of ascitic fluid in abdominal cavity in group C.â‘¡The weight of ascitic fluid in group C were significantly lower compared with those in group B at two time points (P<0.05).3. At two time points, the levels of serum amylase in group B were obviously higher than those of group A (P<0.05); The levels of serum amylase were significant difference between group B and C at 12h and 24h (P<0.05).4. The levels of LPS in serum in group B were significantly higher than those in group A at two time points (P<0.05); In group B,the levels of serum LPS were obviously increased with time prolonged (P<0.05); In group C, the levels of LPS in serum were significantly lower compared with those in group B at two time points. The statistical difference was significant (P<0.05).5. The levels of IL-6 in serum in group B were significantly higher than those in group A at two time points (P<0.05); In group B,the levels of serum serum IL-6 were obviously increased with time prolonged (P<0.05); In group C, the levels of IL-6 in serum were significantly lower compared with those in group B at two time points. The statistical difference was significant (P<0.05).6. At two time points, both the levels of NO in serum in group B were significantly higher than those in group A (P<0.05); In group B,the levels of serum NO were obviously increased with time prolonged (P<0.05); In group C, it was significantly lower compared with those in group B at two time points (P<0.05). There were no significant difference between 12h and 24h in the group C.7.â‘ The levels of MDA in serum in group B were significantly higher than those in group A at two time points (P<0.05); In group C, the levels of MDA in serum were significantly lower compared with those in group B at two time points. The statistical difference was significant (P<0.05).â‘¡The levels of SOD in serum in group B were significantly lower than those in group A at two time points (P<0.05); In group C, the levels of SOD in serum were elevated compared with those in group B at two time points. The statistical difference was significant (P<0.05).Conclusions: Through the experimental study in rats with severe acute pancreatitis, it confirmed severe acute pancreatitis can cause inflammatory factor elevated, neutrophil excessive oxidative stress, intestinal endotoxemia, and further increase the pancreatic tissue injury and immune function disorders. The experimental results show that xue bi jing can antagonize endotoxin, lower plasma NO, reduce inflammatory cytokines and enhance the body of oxygen free radical scavenging capacity, reduce pancreatic tissue damage.
