Objective To investigate the possibility of the anti-inflammatory effect and curative effect by using hyperbaric oxygen therapy on mice with acute lung injury induced by lipopolysaccharide.And further investigate the mechanism and provide a theoretical proof for clinical application of hyperbaric oxygen treatment.Methods All the 46 Sprague-Dawley rats were randomly divided into four groups:normal saline group with 10 rats, LPS group, HBO-A group and HBO-B group with 12 rats. LPS was injected via the tail vein with 5mg/kg.Then arterial cannulation in carotid artery for these rats and at 3 hour,6 hour, respectively, leucocyte count, arterial blood gas analysis,interleukin-8 in blood and lung wet/dry weight were compared. Meanwhile pathological changes were examined under optical microscope.Results①Compared with NS group, the rars of LPS group was less activities and accelerated breathing and distress of respiratory.And there were many areas with inflammatory cell infiltrating of the lung pathological sections of the LPS group.②Compared with blood gas analysis of NS group,the PaO2 and pH were dropped in some extent. The PaO2 of HBO-B group was higher then LPS group (P<0.05) and the pH wasn't variability in the two groups(P>0.05).And there weren't variability between HBO-A group and LPS(3h) group.③Compared with NS group,the lung wet/dry weight was bigger in LPS group,HBO-A group and HBO-B group.And LPS grpoup was the biggest in the three group(P<0.05).④HBO-A group compared with LPS(3h) group and HBO-B group compared with LPS(6h) group, the neutrophil were decrease in HBO-A group and HBO-B group.And the variability was bigger in HBO-B group(P<0.05).⑤The IL-8 of NS group was the smallest. HBO-A group compared with LPS(3h) group and HBO-B group compared with LPS(6h) group,the variability of IL-8 was significant(P<0.05).⑥To be seen on the lung histopatheologic slides,the inflammatory cell infiltration was lessened in HBO-A group compared with LPS group and HBO-B group was the best one in the three groups.Conclusion The HBO therapy could increase the OPP and improve oxygen deficiency and reduce pulmonary edema with increasing supply of oxygen for the tissues and cells and have anti-inflammatory effect, as a result it might have a protective effect against ALI.
